Objective: To investigate the drug resistance phenotype and distribution of serotype of E.coli from chicken in some parts of Shandong Province. Meanwhile,to study different extracts of Folium Artemisiae Argyi,Rhizoma Cimicifugae elimination effect to ciprofloxacin resistant E.coli. In order to reduce the spread of drug resistance and increase the comprehensive utilization of traditional Chineses medicine in a further way.Methods: Using differential medium, Gram stain, biochemical identification and other methods to separate and identify 96 strains of Escherichia coli from Jinan, Weifang, Yantai, Dezhou, Jining of Shandong in 2008-2012. And using the K-B Method to test the bacterical strain’s drug resistant to 23 kinds of usual antibiotics,to know the bacterial strain’s drug resistance phenotype.Through the macroscopic agglutination test to define the serotype of bacterical strains.Adopting the tube dilution method to test ciprofloacin’s MIC to 30 strains of resistance E.coli, then choosing the strains which show the strongest resistance to the CIP and artificial induced resistance strains as the tested strains. Chooseing Folium Artemisiae Argyi,Rhizoma Cimicifugae with good antibacterial effect in vivo and in vito as the drug resistance eliminating agent. Obseving the Chinese traditional medicine eliminating agent’s effect of drug resistance elimination to the tested strains which are set 1/2 MIC,1/4 MIC,1/8 MIC experimental time as 24 h and 48 h. Employing the replica plating to observe drug resistance elimination factor and the changing of the drug resistance phenotype and extract the tested strains’ plasmid,then conduct the agarose gel electrophoresis and observe the different extracts of Folium Artemisiae Argyi,Rhizoma Cimicifugae eliminating effect to the plasmid of resistance E.coli with CIP. Through the plasmid tranformation resistance screening to study the strains’ resistance mechanism in a preliminary way. This test is using the SDS as the positive control, the strains propagating 10 generations natrually as the negative control, and different extracts of Chinese traditional medicine’s eliminating effect as the contrast.Results: Investigate resistance phenotype of chicken E.coli from the parts of Shandong Province,results show that E.coli indicates different and multiple drug resistance to 23 kinds of antibiotics in which it has serious drug resistance to the quinolone.As to CIP, the drug resistance rate reaches to 77.08%. And in different places, the drug resistance of strains in different years is still going on. Ninety-six strains of E.coli are distributed in 024, 015, 018 which are the most common serotype in some places of Shandong Province.According to the selecting results of single traditional Chinese medicine that cures the E.c oli in chicken, it shows that Folium Artemisiae Argyi, Rhizoma Cimicifugae extracts have effective treatment. They are the Chinese traditional medicine eliminating agent of this experiment. The result of artificial induction to standard strains’ drug resistance indicates that the CIP’s MIC to standard strains is up to 10 μg/ml.Different extracts of Folium Artemisiae Argyi,Rhizoma Cimicifugae to tested strains are set in1/2 MIC, 1/4 MIC, 1/8 MIC and 24 h, 48 h accordingly. The effect of Chinese traditional medicine eliminating agent to tested strains is not clear and not in the range of drug resistance. It does not recove the sensibility of strains. The extracts of Folium Artemisiae Argyi and Rhizoma Cimicifugae has the effect of eliminating plasmid to different strains. But according to the result of plasmid transformation test, the drug resistance of tested strains is not certainly decided by plasmid-mediated. It reminds us that the mian way which decides the drug resistance elimination has no direct relationship with plasmid probably. It may be controlled by the other eliminate mechanism.Conclusion: The result of this study show that E.coli from the parts of Shandong province have been severe drug resistance,and the experiment reminds that Folium Artemisiae Argyi,Rhizoma Cimicifugae has drug resistance elimination to the strains of resistance CIP in some extent which have great potential as the clinical curing agent to the E.coli.
