Tobacco mosaic virus(Tobacco mosaic virus,TMV) is one of the serious virus diseases which was widely spread in the world and can threaten the tobacco production yield. Unfortunately, there is not efficient method to control this virus. Until now, only prevention-oriented, integrated control method can be applied to face this problem. Therefore, it is very important to explore a effective prevention method to fight against the TMV.Dry mycelium of penicillium chrysogenum (DMP) is the rest byproducts in the production process of the penicillin, which was widely used in the planting of Yunnan tobacco as a organic induced resistance reagents. To explore the prevention and inhibition mechanisms of DMP to TMV, in this study, Nicotiana glutinosa and N. benthamiana were inoculated with purified TMV and agrobacterium-mediated infectious clone of TMV (p35s:30B:GFP).The predominent results of this study are summerized as follows:(1) Nicotiana glutinosa plants were treated by DMP with three different manner: systemic induced, local induced and passivation in vitro. The inhibition rate were acquired by calculating the number of necrotic lesions which indicated that the DMP has a better control effect to TMV. Local induce effect was also observed in this experiment. However, the treatment effect to the infected plants was not significant. and also has the,but the inactivative was not obvious either.(2) Gene expression assay and plants fluorescence detection were used to study the progress of viral movement, he gfp gene expression assay showed that the gfp gene can be detected earlier before the fluorescent signal can be seen under the long-wave length ultraviolet light. The results showed that infected clone can achieve the completely invaded from the inoculated site to the vascular tissue in a short time and the details was listed as follows:The infected clone could move to the root4hours after inoculation, and4.5hours later, it could reach to the developing leaves at the top of the plant.(3) The plants were inoculated with the infected clone of TMV after24hours when the plants were processed with Penicillium inactivated mycelium(DMP).Fluorescence observed that the fourth leaf of the CK group began to appear green fluorescence,but the DMP group were not in the six days post-inoculation.Then the area of the fluorescence in the fourth leaf of the CK group were bigger, and the fluorescence of the DMP group began to be seen, but the other leaves of the two group were no change. The first nine days later.there were not significantly different between the two groups.
