| China is a big tobacco country where tobacco and its products played a significant role in national economy, and research about tobacco disease-resistant mechanism had been carried forward for years. We established a suppression subtractive cDNA library of tobacco induced by Dry Mycelium of Penicillium Chrysogenum(DM), aimed at reserving gene and providing a good platform for researching the mechanism of tobacco inducible resistance.we established a suppression subtractive cDNA library of5-6leaf stage Nicotiana tabacum L. cv. Honghuadajinyuan treated with0.4%DM and water respectively. A positive suppression subtractive cDNA library including356clone and a reverse suppression subtractive cDNA library covering310clone were successfully constructed through experiments. All clone was plump and clear. Through statistical calculation, the titer of the library was2.2×109cfu/ml, and the ratio of positive cloning was86%.77positive cloning was sequenced to determine the DNA sequence, and the average size was298bp. The sequence was compared with NCBI using Blastn and Blastx, got26protein of different function and7unknown function predicted protein. The results also showed a gene with homology sequence but without homology protein, and a gene without homology sequence or protein.There were26protein of different function among77positive cloning, which mostly related to energy metabolism, protein synthesis, eucaryotic cell structure, signal transduction, disease-resistant defense and cell trauma. Among all the clone there was two clone relating to transcription, eleven clone to energy metabolism, two clone to protein synthesis, two clone to signal transduction, nineteen clone to disease-resistant defense and seven clone to cell trauma, with another thirteen unknown function positive clone.Among the47genes known function, the genes related to disease defense reached a ratio of41.3%, accounting for the most. The energy metabolism and disease-resistant defense genes was in the next place, with a ratio of23.4%and14.9%respectively. The genes related to eucaryotic cell structure, protein synthesis, transduction and signal transduction were fewer, with a ratio of8.5%,4.3%,4.3%and4.3%respectively.We established a suppression subtractive cDNA library of tobacco induced by Dry Mycelium of Penicillium Chrysogenum(DM) and carried on sequencing and comparison analysis towards some of the positive cloning, which would play a positive role in researching tobacco resistance genes. |
