Pirifomospora indica was originally isolated from the rhizosphere of the woody shrubs growing in Indian Thar desert in1998by the India scientist Ajit Verma, and the root-colonizing abilities were shown. It colonizes roots of a broad range of plants and confers enhanced growth, and improves resistance against biotic and tolerance to abiotic stress. Brassica napus L. is on the top of the four oil crops in China. Recently, B.napus has received various bitic and abiotic stress from seeds, seeding and to adult stage, this makes a large effect on the rapeseed quality. In this study, we explored the effects this fungi did to the development and quality of B.napus in filed. Meanwhile, we improved the culture condition and medium prescription of P.indica, and made preliminary study about P.indica formulations. Therefore, this study provide basis for the production and commercial application of P.indica. Our study results are as follow:1. The improvement of culture condition and medium prescription of P.indica:To improve P.indica culture condition, We made four improved Kafer media which included nano carbon, nano TiO2nano ZnO or nano Fe2O3The results shows P.indica grows better and provides more biomass when adding some nano carbon or nano ZnO into traditional Kafer medium. P.indica achieved the maximum biomass when the medium contained0.15g/L nano ZnO or0.2g/L nano carbon, they respectively improved31.7%and66.4%. We made sure that P.indica grows best when the temperature is28℃, and the PH is7.0through different temperatures pH gradient tests.2. Influence of improved P.indica culture filtrate on tobacco growth:In the study, we used the improved P.indica culture filtrate which contained nano C and nano ZnO to water tobacco. The result shows that the improved culture filtrate with nano ZnO is better for tobacco growth than the traditional culture filtrate, the number of senescence leaves in tobacco when watered by the improved culture filtrate with nano ZnO decreased by40%than watered by traditional culture filtrate. 3. P.indica formulations exploration:In this study, we fermented P.indica on barley, bran and com meal medium, or mixed P.indica mycelium with talcum powder, then preserved the mixtures in4癈,25癈and normal room temperature condition, and tested the living P.indica in different formulation every3months. The results showed that after three months, talcum powder was best for P.indica preservation25癈temperature, the living P.indica was132.65%to the original in air condition and96.94%in no air condition; the second suitable method was to preserve P. indica using talcum as carrier in4癈, the living P. indica was74.48%to the original in air condition; the third suitable method was to ferment P. indica on barley and then preserved the mixture, the living P.indica was65.44%to the original in air condition and62.50%in no air condition. After sixth months, the effective endophytic fungi dropped sharply in all of the four formulations, approximately less than30%.4. Influence of P.indica on growth and seeds quality in Brassica napus L.Through two consecutive years of field tests, the study results showed that P.indica can obviously enhance growth, and promote B.napus earlier bolting and flowering, and enhance the seeds yield and quality. The promotion affect is much more obvious when carrying experiment under less nutrient soil condition. In normal fertilization and management conditions, P. indica can reduce erucic acid (2.4%) and glucosinolate (18.14%) content in B. napus seed, and improved the rapeseed oil content (11.1%) and Zn content (10.1%). In less nutrient soil condition, P.indica can promoted oil content (7.78%) and mineral elements Ca (1.03%), Mg (3.27%), Fe (47.03%), Zn (8.94%) contents.
