Although in recent years, a lot of research to the mechanism infection,drug treatment and vaccine research for Mycoplasma pneumoniae of swine had been done,but still cannot effectively quickly to the diagnosis and prevention and control forMycoplasma pneumonia of swine, the damage to the industry economy is still huge.The epidemiological investigation,serum epidemiological investigation, screening ofmolecular epidemiology investigation, pathogen isolation medium and the separation ofpathogenic bacteria identification experiment of the Mycoplasma pneumonia of swine wascarried out for some large-scale pig farms in the part of northern xinjiang, proved the northernxinjiang region pig epidemiological features of Mycoplasma pneumonia, scale pig farms innorthern xinjiang area effectively control and prevent outbreaks of swine Mycoplasmalpneumonia provides a scientific basis.1.For the preliminary understanding of pig Mycoplasma pneumonia infection situation innorthern xinjiang region,16large-scale pig farms in five areas of northern xinjiang hascarried on the field investigation and questionnaire survey, at the same time the pathologicalobservation. Results: the pigs in north xinjiang area of Mycoplasma pneumonia mainly occursin fall and winter, all ages were susceptible to swine, high incidence (30%~90%), lowmortality rate (8%~20%); Clinical symptoms of cough, asthma, abdominal breathing, spiritis depressed, loss of appetite, stagnant growth and development, became thinner, thick coat, inchronic cases, course for months; Disease of the lungs present massive pneumonia anddifferent levels of emphysema and consolidation, show "specimens" or "pancreas"; Alveolilymphoid cell infiltration, mixed with monocytes, neutrophils, and loss of the alveolarepithelial cells.2.456copies of serum were collected randomly from16sterile large-scale pig farms inthe five areas of northern xinjiang, using ELISA method to pig serum epidemiologyinvestigation of Mycoplasma pneumonia. The result shows: in the456serum samples, theantibody positive rate of Mycoplasma pneumoniae was50.4%(230/456),16farms infectionpositive rate was68.75%(11/16), the sow antibody positive rate was92.5%(74/80), thegrowing and fattening pigs antibody positive rate was54.54%(66/121), weaning pigletsantibody positive rate was35.29%(90/255), showed that Mycoplasma pneumonia of swine iswidely popular in the xinjiang region of northern xinjiang.3.To understand the north pig molecular epidemiology features of Mycoplasmapneumonia, collected214suspected pulmonary lung sterile from the16large-scale pig farms,PCR method was conducted for the detection, sequencing for the positive products and thehomology analysis had done between the sequence and which in Genbank logged-in standard strains sequence. Results showed that214lesions in the lungs, positive disease expected for122, positive rate was57.01%(122/214), P36gene sequences with the reference strainhomology was97.3%~100%,P97gene sequences with the reference strain homology of98.1%~99.7%. Test results show that in parts of northern xinjiang, the pneumoniamycoplasma of swine infection is serious, sequencing results show that the local epidemicstrains strains had high homology with international standards.4.To understand the Mycoplasma pneumonia culture characteristics, select the optimalculture medium for the growth of Mycoplasma pneumoniae, the color change unit (CCU)method is adopted and polymerase chain reaction (PCR) to compare and teste for the growthof Mycoplasma pneumoniae in four kinds of commonly used medium and improved KM2,and the separate identification of the pathogen were conducted. Results showed that themodified KM2media was the optimum culture medium,10days color change unit of up to108ccu/mL; At the same time using the medium made a separation successfully. |
