Swine fever(SF) is the malignant contagious disease to damages our swine industry. Recent year, for cultivation administration mode changing , production of pork circulating, induced the prevalence of SF was more complicated. So, to utilize molecular biology technology can expeditiously coping the SF urgent examination, simultaneously ,it also can analyze gene sequence of epidemic strain, in order to know the mutation. The E0 protein,for CSFV, was one of the significant envelope protein, changing or deletion His297,His346, histidine in two according site will attenuate virulence of CSFV, even viral replication prohibition. It showed E0 concerned with virulence of CSFV. So according to the Shimen strain sequence on GenBank, chose conservation area, synthesis two pair of primer named E0DF/E0DR E0EF/E0ER.Useing the primer amplificated the E0 gene by nPCR, to detected the sample from about ten province and autonomy district in china, and do the molecular epidemiology investigation.The result :(1) The text according to the Shimen strain sequence on GenBank, synthesized two pair of primer named E0DF/E0DR E0EF/E0ER, and detected its specificity and sensibility. Detecting the sample of lymphaden, spleen, tonsil from about ten province and autonomy district by nPCR, then the result proved that the primer refer to the CSFV detecting.(2) By the nPCR, detecting the 1600 sample from about ten province and autonomy district in china, the masccline SF detection rate was 26 percent in 2005, 39 pencent in 2006.(3) For E0 gene of SF epidemic strain, the nucleic acid variation was from 0.0 percent to 18.0 percent, the amino acid was from 0.0 percent to 19.0 percent, between epidemic strain and reference strain HCLV the nucleic acid variation was from 1.0 percent to 17.0 percent, the amino acid was from 0.4 percent to 18.0 percent. Among those SF epidemic strain, HEN74, LN163, HEB30, AH44, ZJ87, ZJ47, ZJ94, ZJ69, HUN84, SD89, AH76, ZJ73, FJ216, AH24 and the major reference strain Shimen,HCLV,CHVRI in our country belong to gene 1group; the others belonged to gene 2 group. Showing that gene 2 group was the preponderance strain in most provience of our country. To compare the amino acid sequence of the HCLVstrain, variation at active region on 75~82 site in the amino acid sequence of SF epidemic strain only found in GX31, HEB56, HEN74, HEN128, HEN58, HEN173, HUB22, HUB41, ZJ64, AH82, FJ279.The text used nPCR to detect SF infection in about ten province and autonomy district in china, and procured rarest epidemiology information of SF, to establish the foundation of the prevention and cure also the research of epidemiology.
