Main symptom is the disease cough, gasping, progressiveemaciation and pulmonary interstitial hyperplastic inflammation, etc. Mycoplasma pneumonia of sheep it isquite difficult to control and eliminate, because after sheep infected with mycoplasma pneumonia insubacute performance, combined with rehabilitation sheep can also become contaminated with for a longtime, so the disease is a serious loss to the sheep industry in many countries. Xinjiang breeding sheep mostprovinces in China, until the1990s are the reports of the disease, in recent years owing to the introductionand intensive sheep breeding in xinjiang area of development needs, xinjiang outbreak of large-scale sheepmycoplasma pneumonia, make the production and development of animal husbandry in xinjiang received aserious impact.Currently studying DNA methylation and pathogenic infectious diseases rarely reported, from theirown point of view in order to raise sheep for pleuropneumonia resistance to infection, this article optimizethis species suitable for detecting sheep genome-wide methylation methods: MSAP (Methylation SensitiveAmplification Polymorphism) technology to sheep (the lake mainly sheep) as experimental materials,MSAP technology to optimize the key factors in the selection, through different levels to explore factors inthe selection of four pairs of optimization system under primers shark teeth through8%sequencing gelpolyacrylamide gel electrophoresis the DNA methylation patterns of sheep analyzed to obtain a clear andreproducible silver staining pattern. Indicates that the system can be used for sheep DNA methylationanalysis of the correlation experiment.Through the MSAP (polymorphism) methylation sensitivity experiment method, optimization, to20head sick sheep and20normal sheep genome DNA methylation and infectious pleuropneumonia relevancepreliminary analysis experiment. In sheep in infectious pleuropneumonia ill before and after the wholegenome DNA as the research object, through the crack with enzyme HpaII and MspI to double enzyme, atlast, by sequencing gel polypropylene gel electrophoresis process, from the16MSAP selective primercombinations selected in the preliminary experiment results by clearer4primer combinations for sheep atmethylation changes before and after the assessment. Sick sheep methylation level statistics analysis isgenerally higher than that of the sick sheep methylation levels, methylation polymorphisms at the sametime also will increase.Results show that infectious pleuropneumonia sick or not, and the sheep of the degree of methylationhas certain correlation. The research between epigenetic disease in animals and animal association play acertain reference significance. Speculated that sheep contagious pneumonia with the occurrence anddevelopment of genomic DNA methylation is relevant and can be discussed from the Angle of themethylation host related problems in prevention and treatment of diseases, etc.
