Whipgrass [Hemarthria compressa (L.f.) R. Br.] is perennial grass, belong to subfamily Panicoideae of Poaceae, mainly distributed in tropical and subtropical regions, some also covered in the temperate humid regions of Northern areas. Wild whipgrass’ resources are very rich in China, mainly used in the grassland livestock systems of tropical and subtropical regions. Because of its low seeds production, at present mainly dominated by vegetative propagation. In order to discuss the rapid propagation system of whipgrass, five kinds of whipgrass’young stem was used as explant and using tissue culture technology in this experiment, in order to explore the different genetype whipgrasses in different sample time rapid reproduction system. The main results were as follows:(1)Find the best way of disinfection for the development callus. Young stems of Hemarthria compressa were flushed under the tap water for one hour, then submerged in75%alcohol for10seconds, washed with sterile water3~4times, then immersed in0.1%mercuric chloride for8minutes, washed it again4-5times.(2)Callus induction stage:MS+2,4-D1.5mg/L was the best medium for "Guang Yi"、"Chong Gao" and "Ya’an", callus generation rate of young stem was89.16%、82.09%and86.25%. The explants stems of "T001" and "T002" in the jointing stage, the best medium for stems callus induction was MS+2,4-D3.5mg/L, callus generation rate was89.99%and86.66%,the explants stems of "T001" and "T002" in the heading stage, the best medium was MS+2,4-D1.5mg/L, the generation rate was90.83%and85.83%.(3) Callus subculture culture stages:The highest rate of "Guang Yi" and "Ya’an callus generation happened in the medium of MS+2,4-D1.5mg/L, an d Chong Gao in the medium of medium of MS+2,4-D2.5mg/L.The best culture medium of "T001" and "T002" was MS+2,4-D3.5mg/L. All of them have high quality granular embryogenic callus.(4) Callus differation stage:MS+6-BA1.0mg/L+NAA0.2mg/L was suitable for subculture of "Guang Yi","Ya’an" and "Chong Gao", callus differation rate was76.67%^73.33%and70.00%respectively. The highest rate of callus differation for "T001" and "T002" happened in the medium of MS+6-BA1.5mg/L+NAA0.2mg/L, the rate was76.67%and73.33%。 (5)Regeneration rooting, hardening and transplanting stage:The root formation rate was97.8%in the medium of1/2MS withour any hormone. After7day’s acclimatization, plants were transplanted in cultivation medium (pearlite:muck=2:1) that survival rate was100%.
