Mainly by using Pot culture methodology and started with the systematic analysis of Plant biology and Physiological responses on drought stress, studies on regulation of different Concentrations of exogenous nitric oxide to woad seedlings Resistance and growth and development were carried out in present thesis, including its Possible mechanisms of Plant drought resistance under drought stress in seedlings and were measured the indigo quality of its extraction under different concentrations of SNPS by HPLC.The major results are presented as follows:1.0.2-2mmol/LSNPS at two days could alleviates drought stress. The best effect was observed in the pretreatment of 0.5mmol/LSNP, the effect of 10%PEG6000 with them on the germination rate, germination Potential, germination index was more obvious lower than normal groups,however,0.5mmol/LSNP at two days dark environment could alleviates drought stress. comparing with normal groups, their germination rate, germination Potential, germination index respectively increased 57.90%,44.19% and 56.71%, thus the difference is extremely significant (P<0.05). The relative water content is one of important indexes that reflect moisture conditions in plant. So the water content of seedlings root and leaf is basically uniformity in stress conditions. Meanwhile appropriate NO concentration alleviated the drop of the relative water content of seedling root and leaves under drought stress, and cut down on water loss in plants, even significantly reduced the seedlings damage.2. Under drought stress condition, the different concentrations at 0.2,0.5,1.0mmol/L SNP could alleviates Oxidative damage, and enhanced the enzyme activity of SOD, POD and CAT,0.2,0.5mmol/LSNP seedling made significant effect (P<0.05).And Significantly enhanced the enzyme activity of SOD and POD. When drought stress condition at 1-2d, all enzymes are maximum. But with lengthening the stress time, the activity reduced. The results indicated that SNP had a certain adjustment range.3. Exogenous NO reduced the young seedlings accumulation of Membrane lipid Membrane Lipid Peroxide Product MDA under drought stress, During the drought stress, the content of MDA in drought stress seedling increased with lengthening the stress time, SNPS treatment has similar trend, but relatively slow.because exogenous NO have been membrane phase change, thus eased membrane lipid peroxidation, and the most significant effect about Membrane protection was 0.5mmol/LSNP (P<0.05)4. Exogenous NO significantly induced the accumulation of the Proline and Soluble Sugar (P<0.05) under Drought Stress, in which SNP groups was significantly higher than CK groups, and the differences was most obvious in 0.5mmol/LSNP. The results indicated that exogenous NO could relieve the damage of the young seeding by adjusting Penetration material metabolism under Drought Stress, And that Exogenous NO improved infiltration protection ability of the leaves under drought stress conditions.5. The research suggested that we should adopt HPLC to measure the indigo quality extraction with different concentration SNP, firstly made it volatilize the alcohol, then used the chloroform to refine the indigo, finally detected by HPLC, Using ODS (4.6×100mm,5μm) with mobile phase consisted of methanol-0.1% ammonium acetate-acetic acid(70:30:1), and the UV detection wavelength was set at 280nm, The results indicated that Indigotic has a very linear relationship at 0.00125 mg·ml-1~0.005mg·ml-1 concentrations, r=0.9921. through indigo standard and the HPLC picture of the samples, we concluded that the extraction reached to peak area in the 3-4 minutes in different concentrations SNP, and that the extraction content achieved the highest peak in lmmol/LSNP,that is 0.047832mg/g, RSD=0.31%.
