The genus Trichinella spp. is a food-borne zoonotic parasite with a cosmopolitandistribution. The trichinellosis is caused by humans and animals eating uncooked orsemi-cooked meat with infective worms. The outbreak of disease is still occurred insome developing countries (such as Thailand, Argentina and China, etc.), in additionto the reason of the lack of effective prevention and control measures, the key is stillunfound some good diagnostic methods. The trichinellosis is no typical clinicalsymptom, and it is easy to confuse with other infection, so which makes the clinicaldiagnosis become very difficult. Although muscle biopsy may diagnose, but notofteneasy to be discovered in the mild infection.The biopsy positive rate also onlythen about50%, and it is not accepted easily by the people because of theorganization limited influence. Therefore, the research and development of onesensitivity, specificity sensitivity diagnosis method would be the most importantsignificance for diagnosis, preventing and controlling trichinellosis.The mice as experimental animal models and slaughter pigs applied to clinicaldiagnosis were inoculated with muscle larvae of Trichinella spiralis(T.spiralis,T1)from pig Trichinella isolates (ISS534) in Henan province of China in this study,excretory-secretory antigen (ES) from adult stage (Ad3), Newborn larvae stage(Ad6+NBL),intestinal muscle larvae(10h),intestinal adult(30h) and muscle larvae (ML) ofT.spiraliswere coating antigen, the tail vein blood of infected mice was collected indifferent days within two months and serum was separated, the IgM and IgGanti-Trichinella have been detected by ELISA method, and using Excel software todraw their antibody dynamic curves followed by data analysis. The results indicatedthat anti-Trichinella IgM antibodies in mice serum were detected to change curve inthe form of "peaks and valleys" with three antigens, the mixed ES antigens fromthree developmental stages could be used to detect the Trichinella infection before45days post infection(dpi)(the serum anti-Trichinella IgM antibody was detected in the5-14dpi and anti-Trichinella IgG was detected in the15-45dpi), the ML ES antigen could be used to detect the Trichinella infection after45dpi, the most significantfinding was that Ad3and Ad6+NBL ES antigen could be used in14-19dpi. For pigs,the mixed ES antigens from3-5developmental stages could be used to detect the lowdose Trichinella infection before2month, the10h ES antigen could be used to detectthe serum anti-Trichinella IgM antibody in the middle or high dose Trichinellainfection before60dpi. This study successfully obtained the dynamics ofanti-Trichinella antibodies from the stimulated host by T.spiralis infection anddetermined the ES antigen in different developmental stages of T. Spiralis for thetrichinella immunological diagnosis, which would provide the important experimentalfoundation and theoretical reference value for the establishment of effective diagnosispreparations on Trichinella spp. infection with the different developmental stages ESantigen from Trichinella spp.
