As a rare precious chinese herb, Dendrobium officinale has great value for drug. But it was endangered for its wide application and special growth conditions. Embryoid of Dendrobium Candidium Wall Ex Lindl(ED. Officinale) was obtained through tissue culture techineques, with high reproduction speed and low cost advantages. In order to provide the theoretical basis for the further research and application of ED. Officinale and solve the problem of endangered plant resources in short supply, the present paper was designed to establish the ED. Officinale’s quality specifications and to evaluate the bioactivity of ED. Officinale, including antioxidant, anti-aging and improving sleeping effect.Firstly, through the analysis of ED. Officinale’s sensory index, TLC identification, the content of polysaccharides and alcohol content of different batches, we established the ED. Officinale’s quality specifications. The content of polysaccharides should not less than8%, and the alcohol content should not less than6.5%.Secondly, we studied the the antioxidant effect of ED. Officinale on drosophila. First of all, through the survival test, the biochemical test and the antioxidant enzymes’s gene expressions test, we discussed the effect of ED. Officinal on the lifespan and antioxidant system of drosophila. The results showed that ED. Officinal could prolong the lifespan of drosophila, incresase the.activity of SOD and CAT and decrease the LF’s content. Real-time PCR amplification assay was carried out to quantify the antioxidant system’s mRNA level, the results showed that mRNA gene expressions of Cu,Zn-SOD, Mn-SOD and CAT of drosophila feeding ED. Officinal were increased compared with those of control group. Besides, the anti-aging effect of ED. Officinal in drosophila damaged by UV and D-galactose were studied. The results showed that compared with the model group, the drosophila’s life of dose group was prolonged, the activity of antioxidant system was increased and the LF’s content was decreased, indicating that ED. Officinal had antagonistic action on the damage induced by UV and D-galactose. At last, hydrogen peroxide treatment and paraquat treatment were used to examine the resistance of drosophila against oxidantive stress. The results showed that ED. Officinal could prolong the average lifespan and maximum lifespan of drosophila.Thirdly, through the sleep-extending test with sodium pentobarbital, the sleep-producing test with subthreshold dose of sodium pentobarbital, and the sleep incubation period test with sodium barbital, we studied the ED. Officinal’s sleeping effect on mice. The results showed that ED. Officinal could prolong sleeping time with sodium pentobarbital, increase sleeping rate under the threshold hypnogenesis dosage and shorten the incubation period induced by sodium barbital, indicating that ED. Officinal had improvement effect on sleep. The results above suggest that ED. Officinal shows obvious antioxidant activity and improving sleeping effect.
