Curcumin, orange yellow crystalline powder, is the active ingredient which is extracted from zingiberaceae, araceae plant root and belongs to the plant protection material. It is an exhibits strong antioxidant and anticancer activities; however, low bioavailability restricte its application in chemotherap-eutic trials. In order to increase the natural antibacterial, antitumor compounds and other biological pharmacological activity, appropriate modification was used to improve its applicability. This way has become one of the new methods to research antibacterial and antitumor drugs.In this paper, a kind of curcumin analogues2E.6E-2-(1H-indol-3-yl) methyle-ne)-6-(4-hydroxy-3-methoxy benzylidene)-cyclohexanone (IHCH) as the research object aimed to investigate,yet no study on the resistance of IHCH to plant fungal and cancer.In order to detect IHCH antibacterial and antitumor effect and mechanism, we choose different concentration IHCH (1.1、2.2、4.3、8.5.17mmol/L) of the IHCH processingcuc umber fusarium wilt and A549cells (human non-small lung epithelial carcinoma cancer cells),48h, by determined by MTT, acridine orange staining, MDC staining analysis, autophagy inhibition Wortmannin, LC3positioning, and Western bloting, analysis its antitumor effect and mechanism.The results show that curcumin can significantly suppress the growth of the cucumber fusarium wilt, bacteriostatic effect is concentration dependent. But IHCH can faint restrain the growth of the cucumber fusarium wilt, illustrates the reconstructing lowered its bacteriostatic effect. IHCH can inhibit the growth of A549cells and the inhibition effect show dose and time dependent. By IHCH A549cell morphology changed, IHCH drug dealing with3h A549cells, with the increase of drug concentration, cells developed cavitation in drug concentration20μmol/L processing3h can clearly see the cavitation.40μmol/L cavitation reduced; IHCH36h concentration20μmol/L processing cells, compared with the control cell survival rate significantly decreased; Curcumin analogues (IHCH) treatment after A549cells respectively by acridine orange staining, MDC staining3h, the results showed that20μmol/L, cavitation increase,40μL significantly reduced; LC3positioning results show that the IHCH3h,20umol/L processing A549cells appear obvious LC3dots; Western bloting electrophoresis appraisal of IHCH when handling A549cell3h,20μmol/L, LC3-II/LC3-I,can IHCH inducing autophagy of A549cells.
