In the study, bacterial in milk samples which got from mastitis cows were isolatedand identified in lab. Milk samples were uniformly smeared on the Maconkey Agar, theEosin Methylene Blue Medium and Sheep Blood Nutrition Agar Plates respectively, thenobserving the specific colonies under microscope. The colonies suspected as E. coli wereinoculated on the eosin methylene blue agar plate and the suspected Staphylococcusaureus and Streptococcus Agalactiae on the sheep blood nutrition agar plates respectively.The suspected bacterial were identified through biochemical and found that E. Coli,Staphylococcus aureus and Streptococcus Agalactiae were the main pathogens of themilk.Anti-bacterial function of Di Yu, Jin Yin Hua, Yin Hua and Huang Qin on the threemain cow mastitis were tested through serial dilution method on solid medium, andfound that E. coli and Streptococcus Agalactiae were moderately sensitive to Huang Qinand Di Yu, Staphylococcus aureus was hightly sensitive to Di Yu and Huang Qin.Based on in vitro anti-bacterial test, combining with the price of herb and whethereasy to be made of preparation, Di Yu was chosen to be made of Chinese traditionalmedicine membrane according to certain technology.In order to test the security of the Chinese traditional medicine membrane, duringthe study, skin irritation test and skin allergy test were made and confirmed that themembrance had no irritation or sensitization on rabbits intact skin or damaged skin, andthe irritation index was less than0.5, the allergy index was0. It indicated that theChinese traditional medicine membrane was safe without toxicity and allergic. Toevaluate the safty of the Chinese traditional medicine membrane in milk, mice werechosen to make acute toxicity and chronic toxicity tests. Acute toxicity test showed thatno mice dead after gavaged100mL/kg BW. Based on the acute toxicity grading “HealthFood Safety Toxicity Evaluation Procedures and Methods”, the medicine had no toxicityif the LD50of the medicine more than21.50g/kg BW. There were no significantlydifferences between the clinical observation, organ cosfficients, blood routine tests, bloodbiochemical parameters and histopathological, it indicated that the preparation was safteven long time in milk. To further evaluate the clinical efficiency of the Chinese traditional medicinemembrane on mastitis, lactation rabbits were selected as experimental model which weredivided into3groups as no medicine control group, treatment group and control group.In the treatment group, nipples of rabbit were smeared the Chinese traditional medicinemembrane and then coated with bacterial after milking, in no medicine control group,nipples were coated with bacterial directly, while nipples were smeared normal saline incontrol group. After1week treatment, rabbits in no medicine control group werelistlessness, back hair messy, nipples swelling significantly, milk obvious changingyellow and thickening, accompanied with flocculent. However, there were no abnormalof nipplies in the treatment group and control group. It showed that the Chinesetraditional medicine membrane had protective effect on rabbit mastitis.Anti-oxidant indexes were detected in rabbit blood after the test above finished.Results showed that the level of SOD and GSH-Px form no medicine control group weredecreased significantly compared with control group, while the contents of NO and MDAincreased significantly. Indexes in treatment group showed no significant difference withcontrol group. Results indicated that the Chinese traditional medicine membrane hadprotective effect on rabbit mastitis and then protected the anti-oxidant function of body.
