Sogatella furcifera belongs to Homoptera, Delphacidae, is one of the major pests of rice, which stops the normal growth of rice by sucking sap and spread the virus. using chemical pesticides is the main control measures, but long-term use can lead to the growing of drug resistance and the destruction of natural enemies.Therefore, the most effective measures to prevent Sogatella furciferai is identifying new source of resistance Sogatella furcifera from the wild rice species and transfering new resistance Sogatella furcifera gene,excavated from resistance Sogatella furcifera material, into cultivated rice. This will not only prevent spread of virus, it can also block the Sogatella furcifera direct feeding damage.In this study, we were screening source resistance about 218 Varieties of Guangxi common wild rice in researcher Li Rong-bai seed bank in College of Agriculture, Guangxi University by using Seedling mortality and Seedling stage group methods and part of more resistance antigens from first screening for the second screening; Meanwhile make resistance evaluation, genetic analysis, the initial position of resistance genes of resistant by CL37 and CL32 of rice resistant material Hybrid infected Sogatella furcifera rice material 9311. This study are as follows:1.Select 42 resistant materials from 158 Guangxi common wild rice by seedling mortality, these materials are 26.58% of total identification of materials; Identify different kinds of resistant material from 60 Guangxi common wild rice by Seedling stage group method, in which 1 of 60 for high resistance,21 of 60 for middle resistance; Reselecting the more resistance materials in the first selection of the three Guangxi common wild rice 05WRBPH23,05WRBPH2ã€05WRBPH22, it turns out that the three meterals are all middle resistance.2. Evaluate resistant materials CL37 and CL32 by using The hatching rate of seedlingã€Honeydew amount of Seedling Stageã€Honeydew of mature periodã€Seedling stage group method;three of them shows Different levels of resistance in the four indicators,they are Hatching rate of seeding Honeydew amount of Seedling Stage〠Honeydew of mature period. Seedling stage group method indicate Middle resistance.3. Hybrid infected Sogatella furcifera material 9311and CL37ã€CL32 produces F1 Generation,make a phenotypic analysis of classical genetics of two F2 Generations,which produced by mating with themselves. It turns out that the proportion of susceptible plants to resistance plants is 3:1 in group F2 Generations,which produced by the hybrid of 9311/CL37and 9311/CL32.These shows that CL37and CL32 material contains recessive resistance gene, whose resistance is controlled by a pair of main recessive gene separately.4. Initial locate the two main recessive gene in group F2 of 9311/CL37ã€F2 of 9311/CL32.First, use the Honeydew amount per plant as Phenotypic value in group F2 of 9311/CL37ã€F2 of 9311/CL32; Then, select the Polymorphism of the the resistant and susceptible pool built by the two different groups by using 756 Primers in the whole covered gene group and find The polymorphic primers; Finally, mark SSR in group F2 of 9311/CL37,Find a new site in no.3 chromosome, whose site is between interval RM3526 and RM3513, the LOD is 3.2 and this section’s refered physical distance is 527kb in Nipponbare genome; meanwhile,find a new site in no.3 chromosome, whose site is between interval RM13650 and RM530, the LOD is 3.84 and this section’s refered physical distance is 495kb in Nipponbare genome. |
