Mongolian horse is a famous local breed of China, and has various and excellent genetic resources. It has recorded in the conservation list. Compared with other breeds, the Mongolian horse have unique characteristics resistance of crude feed, fatten rapidly and losing weight slowly. This is also the Viability of Mongolian horse that survived under the harsh environment for long time.Resistance of crude feed characteristic of Mongolianhorse except for genetic factors, also is related to its special intestinal microflora inevitably. In order to understand its microbial functioning relationship, we conduct a preliminary study on the diversity of Mongolian horse and Thoroughbred enteric bacteria. Also provide basis for mining and utilization of functional bacteria.The fresh feces of Thoroughbred and Mongolian horse under the different feeding conditions were used in this study, then isolated bacterial genome DNA from fecal samples with biological kit directly.using PCR technique amplified 16S rDNA V3 area sequences from bacterial total DNA and degeneration gradient gel electrophoresis separated the sequence of different bacteria. Using the Quantity One software to analyze plastic figure, At the same time rubber cutting recycling edge banding and proceed cloning sequencing. Blast the obtained Sequence in the gene database and judge the strain characteristic. The study results are as follows.First, In the comparison of Mongolian horse and Thoroughbred intestinal bacterial community structure, DGGE gel figure showed 53 stripes with different locations. Among the 41 retrieved dominant stripes identification, only 16 clones similarity more than 96% with bacterial 16 s rDNA V3 area in NCBI database. Comparison of structural changes of intestinal bacteria of Mongolian horse from two kinds of breeding condition, DGGE gel figure showed total 50 stripes, selected 40 dominant sequence resulted in 17 clones similarity more than 96%.It indicates low homology. In the two experiments unreported advantageous bacteria posessed 61% and 58% of total bacteria respectively.Second, From stripe number and community diversity on DGGE figure with QuantityOne software, intestinal advantage bacteria of Thoroughbred are more various than Mongolian horse. Cluster analysis result showed Mongolian horse and Thoroughbred get into a brand and intestinal advantage bacterium similarity was only 0.33 between breeds. The similarity within breed is greater than the similarity across the breed. Intestinal advantage bacterium of Mongolian horse that grazing on grassland in July are more abundant than captive horse in march. Mongolian horse after clustering analysis in March and July, Cluster analysis showed they become a brand respectively and middle similarity is 0.54. Within breed similarity of Mongolian horse feed with hay is greater than feed with green grass.Third, Common intestinal advantage bacteria in Mongolian horse and Thoroughbred are Prevotella ruminicola、Butyrivibrio fibrisolvens、Corynebacterium flavescens、Ruminococcus flavefaciens and Bacillus subtilis.all above bacteria have biodegradation fiber function except for bacillus subtilis. Bacillus subtilis is a multifunctional probiotics. Specific intestinal advantage bacteria in Mongolian horseare Ruminococcus sp、Clostridium cellulolyticum、Clostridium saccharolyticum. The former two of them are strong fiber degrading bacteria. Unique intestinal advantage bacteria in Thoroughbred are Clostridium barati、Clostridium aminophilum、Eubacterium oxidoreducens、Ruminococcus gnavus、Clostridium symbiosum、Bifidobacterium lactis、Streptococcus bovis、Bacillus anthracis. Mongolian horse grazing on grassland in July compared with captive conditions in March, new increasing intestinal advantage bacteria are Acetivibrio cellulolyticus、 Eubacterium rectale、Clostridium clostridioforme、Coprococcus eutactus Eubacterium brachy. Onthe contrary, Corynebacterium flavescens and Bacillus subtilis of Mongolian horses that grazing green grass become non-dominant stripe. Intestinal main fiber bacteria species have little change in Mongolian horse under two different breeding conditions.
