Research And Development Of A Probiotics Combined With Chinese Herbal Medicine And Its Application In Tilipia(Oreochromis.spp)

In recent years, the outbreak of all kinds of disease has seriously hindered the development of aquaculture in our country, the use of antibiotics and other chemical drugs had seriously damaged to human health and aquatic animals. As substitutes of antibiotics,Chinese herbal medicine and probiotics of research is on the hot in recent research. On the basis of the synergistic effect of probiotics and Chinese herbal medicine,in this study, they were combined together to use in the tilipia. The results as follow:1. According to the preliminary study of the our laboratory, Chinese herbal compound(A, B, C) were mixed by nine kinds of Chinese herbal medicine, which have proliferative effect on five kinds of probiotics such as Enterococcus faecalis and Lactobacillus acidophilus. The proliferative effect of probiotics on Chinese herbal compound(A, B, C) and the favorable concentration were determined. The result indicated that three kinds of Chinese herbal compound can promote the growth of probiotic bacteria in vitro and the best one is B. The extract of 5% of B has the best effect on Enterococcus faecalis 、 Lactobacillus acidophilus and Bacillus natto, and the best concentration of Bacillus subtilis and Bacillus licheniformis is 8%.2. Four hundred and fifty tilipia(Oreochromis niloticus) of the average weight of(9.5士0.5g)were randomly divided into 5 groups. Control group A was fed with basal diet,treatment group B、D were fed with basal diet which added probiotics, group C、E were fed with basal diet which added probiotics combined with Chinese herbal medicine. At the termination of feeding trial, the effects of probiotics combined with Chinese herbal medicine on the growth performance, intestinal microflora and disease resistance of GIFT strain of Nile tilapia were investigated. The results showed that the addition of probiotics combined with Chinese herbal medicine in feed,(1)the number of intestinal bacteria,Bifidobacterium, Lactobacillus and Bacillus were significantly higher than the control group(P<0.05), the content of E. coli was significantly lower(P<0.05). In addition,compared with probiotics group, the effect of probiotics combined with Chinese herbal medicine is better.(2) There is significant effect on the weight-gaining, specific growth rate(P<0.05), but compared with control group, different treatments had no significant difference on feed conversion( P ﹥ 0.05).(3) After challenged with Streptococcus agalactiae, the immune protection rate of GIFT Strain of Nile Tilapia of differenttreatments was 44.4%、55.6%、55.6% and 66.7%. Probiotics combined with Chinese herbal medicine added into the basal feed could improve the immune protection rate and the proper amount is 2×107cfu/g.Conclusion: Chinese herbal compound(B) can promote the proliferation of five probiotics bacteria in vitro. The use of probiotics combined with Chinese herbal medicine could improve intestinal microflora and had a good promotion effect on the growth and disease resistance of GIFT strain of Nile tilapia.
