Torreya grandis is a unique economic tree species in China, which has very high edible and medicinal value. Semen Torreya is rich in oil with a high content of unsaturated fatty acid and has certain physiological activity,so it is worthy of development and utilization of senior vegetable oil source. Torreya grandis aril,as the processing by-products of Torreya grandis,contains a variety of biological active ingredients.,is also a kind of worthy of exploitation and utilization of sources of natural products. This experiment is study on the extraction and analysis of the main ingredient of Torreya grandis aril and Semen Torreya and their antioxidant activity,the purpose is providing certain theoretical basis to further processing of the resources of Torreya grandis.1. Using the tube and TLC qualitatively identificating the active substance of Torreya grandis aril. The results show that there are amino acids, polysaccharides, saponins, flavonoids, phenols, alkaloids, tannins> and coumarin in Torreya grandis aril.2. Using Ultrasonic assisted extraction to extract the total flavonoid of Torreya grandis aril, through the single factor test and Orthogonal test to study the effect of ethanol concentration, solid-liquid ratio, extraction temperature and extraction time on the extraction yield of the total flavonoid.The results indicated that the affecting of the extraction was the concentration of ethanol> the ratio of material to liquid> extracting time> extracting temperature. The optimal extraction conditions were as follows:ethanol concentration was 60%,he ratio of material to liquid was 1:30(g/mL), extracting temperature was 55℃,extracting time was 35 min,the extraction yield of falconoid reached mg/g under this process.3. Determining the clearance ability of the the total flavonoid of Torreya grandis aril to DPPH free radicals, hydroxyl free radicals and super oxygen anion free radicals, and the results were compared with VC. The results indicated that the total flavonoid of Torreya grandis aril could effectively scavenge the three kinds of free radicals,which is equal or superior to VC at the same concentration. Half of the clearance(IC50) is 64.3μg/mL,42.4μg/mL and 186.7μg/mL respectively.4. Using the Soxhlet extraction to extrat the Semen Torreya oil, through the single factor test and Orthogonal test to study the effect of extraction solvent, and liquid ratio, extraction time and extraction temperature on the yield of Semen Torreya oil. The results indicated that the best extraction solvent is n-hexane. The affecting of the extraction was the ratio of material to liquid> extracting time> extracting temperature.The optimal extraction conditions were as follows:the ratio of material to liquid was 1:9(g/mL), extracting time was 4h,extracting temperature was 60℃,the content of falconoid reached 48.05% under this process.5. Determining some of the physical and chemical properties of the Semen Torreya oil and its antioxidant activity. The acid value, iodine value, saponification value and peroxide value is 0.86 mgKOH/g,118.65 gI2/100g,172.42 mgKOH/g and 4.55 mmol/kg The Semen Torreya oil could scavenge the DPPH free radicals and hydroxyl free radicals, half of the clearance(ICso) is 4.47mg/mL and 2.26mg/mL,but it could not effectively scavenge the super oxygen anion free radicals.
