Taxonomy of Lophodermium on pines in Shannxi and Hubei provinces and the genus’s phylogenetics based on two-gene sequences were studied by modern fungal taxonomic method, and 7 species of Lophodermium on pines were found, respectively Leptostroma magnum, Leptostroma coalescens, Lophodermium pinastri, Lophodermium kumaunicum, Lophodermium nitens, Lophodermium cedrinum and Lophodermium himalayense. Among them Leptostroma magnum and Leptostroma coalescens were new anamorphic fungi which always attach to Lophodermium while their teleomorphs existing. All type specimens were deposited in the Reference Collection of Forest Fungi of Anhui Agricultural University, China (AAUF). The research content mainly includes the following two aspects.Firstly, relatively systematic taxonomy of Lophodermium on pines was mainly studied using the traditional taxonomic method. Two new species were preliminarily established by external examination and internal microexamination of frozen section, detail description of morphological features, internal structure characteristics and so on in both English and Chinese were given, and the point line composite image of conidiogenous cells, conidiophores and trichogynes were given. At the same time, the authors also investigated and studied in geographic distribution, ecological habitat of these species.Secondly, combining the molecular biology and bioinformatics methods, based on rDNA-ITS,28S rDNA sequence and two-gene sequences, the phylogenetic trees were constructed to explore the interspecies and intraspecies relationships of the Lophodermium members from Shannxi and Hubei provinces. Total DNA were extracted by CTAB method, the correlation sequences were amplified by PCR test, blasted them in GenBank and selected Leotia lubrica Fr. of Helotiales as outgroup, then phylogenetic trees were constructed respectively to identify the species classification status combining with morphological classification method. The results showed that the same species of different strains usually preferred to cluster together, or different species with the same region and host are also likely to gather together. No matter the phylogenetic trees based on rDNA-ITS,28S rDNA sequences or two-gene sequences, all strains of Lophodermium on pines were gathered in Clade â… , and strains of Lophodermium on hardwood generally gathered in Clade â…¡ According to objective factors existing the traditional taxonomic methods, the results obtained by traditional taxonomy method (morphological identification) and modern molecular biology methods (construct phylogenetic tree) are not identical. In conclusion, just relying on morphology methods to identify fungi is inadvisable, and we should combine method of comprehensive analysis on several characters and modern molecular biology technology, such as combining with fungal morphology, cultivate character, individual development, host, habitat and geographical distribution, and take modern molecular biology techniques as the auxiliary means, only that reasonable and scientific research results could be attained. |
