Lipoprotein lipase (LPL) plays a major role in the metabolism and transport of lipids. LPL gene has been seen as a candidate gene affecting intramuscular fat content. In order to obtain marker loci about goat muscle fat traits and improve the ability of fat deposition, six goat breeds selected were Anhui white goats, Boer goats, horse goat, Jining Grey goats, minshan black fur sheep, and LinQu goats. The six populations were regarded as experimental subjucts. The polymorphisms of LPL were examined using PCR-SSCP, analyzed by using GLM model and to analyse the relationship between the polymorphisms and intramuscular fat content in goats.1.The specific primers p1-p10 were designed according to the relevant goat LPL gene sequences published in GenBank. The P1 polymorphic loci has been found in the LPLgene using PCR-SSCP, with five genotypes AA、 BB、 AB、AC and BC on the site. The measurement results show that the P1 variation is replaced by the nucleotide G'287A. The P6 polymorphic loci has been found in the LPLgene using PCR-SSCP, with three genotypes AA、BB and AB on the site. The measurement results show that the P6 variation is replaced by the nucleotide G'16997A, resulting in Gln'Ser. Three genotypes AA、BB and AB in Exon 7. The measurement results show that the P7 variation is replaced by the nucleotide A'18313C, resulting in Asp'Ala.2.To analyse the genotypes distribution of PCR-SSCP loci in the six groups, the test results show that all populations, in additon to Anhui white goat, have reached the Hardy-Weinberg equilibrium on the site P1 (P>0.05); and all groups have reached the Hardy-Weinberg equilibrium on the site P6 and P7 (P>0.05).3.The SNP analysis of Group Dynamics show,287G、18313 A were stable distribution. The distribution of 16997G has a specific in six populations,with Anhui White Goat 0.9750, Boer goats 0.8828, Matou goat 0.8333, Linqu goat 0.7500 Qu Jining Grey goats 0.7037, and the minimum distribution rate of minshan black fur goats is 0.6979. The G-16997A is negatively correlated with IMF; A-18313C is significantly associated with IMF (P<0.05). It follows that A-18313C is correlated with fat deposition ability.4.According to DA genetic distance, the population heterosis has been predicted that Boer goats and Anhui white goats, horse goat, Jining Grey goats, minshan black fur sheep, and LinQu goats will have the largest heterosis.
