| This study chose seven swinery of Duroc,Landrace,Large White,Jihebai A,Jihebai B, Liaoning black,Dan Yu black a total of 281 pig as the research object,designed primers with pig L-FABPgene sequence which have published in GenBank,PCR amplification and sequencing analyzed.The results showed:intercomparison sequence from Exonl to part Intronl fragment amplification by primers 1 of two individual selection from each swinery with pig L-FABP gene sequence published in GenBank,found that base site of 237,258,315 of all individuals were A,T and A respectively,different with DQ182323 base sequence of G,C and C,and existence G403A mutation between individual. Intercomparison.Sequence from Intronl to Exon2 fragment amplification by primers 2 of two individual selection from each swinery with pig L-FABP gene sequence published in GenBank,found 7 polymorphism loci existence in Individual.Among them,G1546T,C1715T and T1741C three polymorphism loci occurred in Intronl,the other C1755T,T1870C,A1875C和C1897A four polymorphism loci occurred in Exon2,C1755T,T1870C,A1875C were synonymous mutation,and C1897A lead to Leu-Met changed.In G1546T site G allele was dominant in all seven breed.Jihebai B only exists GG genotype,TT genotype weren't detected in Duroc and Jihebai A.Genotype distribution tested by x~2 test showed that there was no difference between Jihebai A and Jihebai B,and between Liaoning black and Danyu black.In C1715T site C allele was dominant except Liaoning black which C/T allele frequency were equation,Jihebai B only exists GG genotype,TT genotype weren't detected in Duroc and Jihebai A.Genotype distribution tested by x~2 test showed that there was significant difference or very significant difference between Jihebai A and the other six breeds.In T1741C site T allele was dominant except Liaoning black which C allele was dominant,C allele frequency of Liaoning black,Dan Yu black,jihebai A,jihebai B,Duroc,Landrace and Large White were 0.781,0.643,0.152, 0.651,0.755,0.718,0.419 respectively.Genotype distribution tested by x~2 test showed that there was very significant difference between Large White and the other six breeds (P<0.01);there was significant difference or very significant difference between Dan Yu black and the other six breeds.In C1755T site C allele was dominant in all seven breed.Duroc,Liaoning black and Dan Yu black only exists CC genotype.Genotype distribution tested by x~2 test showed that there was significant difference between Landrace and the other six breeds.;there was no difference between Liaoning black,Dan Yu black and Duroc,in this site C allele was correlation with intramuscular fat content.Y1870C,A1875C,C1897A three sites showed linkage,in T1870C site T allele was dominant except Landrace which C allele was dominant,Duroc T allele frequency was highest(0.875),the CC genotype was not detected.In A1875C site A allele was dominant except Landrace which C allele was dominant,Duroc A allele frequency was highest(0.875),the CC genotype was not detected. In C1897A site C allele was dominant except Landrace which A allele was dominant, Duroc C allele frequency was highest(0.875),the AA genotype was not detected. Genotype distribution of three mutation site tested by x~2 test showed that there was significant difference or very significant difference between Landrace and the other six breeds.There was no difference between Liaoning black,Jihebai B and Danyu black,there was significant difference or very significant difference between Liaoning black and the other four breeds.In the L-FABP gene seven mutation site,polymorphic information content G1546T, C1715T sites PIC content of Liaoning black was high,as highly polymorphic,Landrace and Danyu black were moderately polymorphic,and Jihebai B were the single state,the other breeds PIC content was lower,as low polymorphism.In T1741C site PIC content of Landrace,Large White,jihebai A and Dan Yu black was high,as highly polymorphic,the other breeds were moderately polymorphic.In C1755T site Landrace and jihebai B were highly polymorphic and moderately polymorphic respectively,Duroc and Liaoning black were the single state,the other breeds PIC content was lower,as low polymorphism.In T1870C site PIC content of all seven breeds were high,as highly polymorphic.In A1875C and C1897A sites PIC content of Landrace and Liaoning black were high,as highly polymorphic,and Duroc was low polymorphism,the other four breeds were moderately polymorphic.The results showed that there was significant difference among breeds in IMF content except between Jihebai A and Duroc.IMF content of Liaoning black,Dan Yu black,jihebai B,jihebai A and Duroc were high,next was Landrace,and Large White IMF content was low,there were varieties among breeds in IMF content.There was no significant difference in IMF content of Genotype of G1546T,C1715T and T1741C three mutation sites in intron. In C1755T site of Exon,IMF content of CC genotype was significantly higher than the CT genotype and the TT genotype;T1870C,A1875C,C1897A three sites showed Linkage, there was no significant difference among genotype in IMF content,in T1870C site existence genotype TT>CT>CC trend in IMF content,in A1875C site existence genotype genotype AA>AC>CC trend in IMF content,in C1897A site existence genotype genotype CC>AC>AA trend in IMF content,IMF content of three sites TT-AA-CC genotype was higher.Clustering results show that jihebai A combined with jihebai B first,and then combined with the Large White,with the Liaoning Black and Tanyu Black together; combined with Duroc and Landrace in turn.Indicate that there was close relatives between Jihebai A and Jihebai B,Liaoning Black and Tanyu Black.In this test we analysis L-FABP sequence polymorphism,to discover new polymorphic site on L-FABP,study the relationship between different genotypes of L-FABP and intramuscular fat(IMF)content,there by to lay a foundation for the determination of the major gene that effects intramuscular fat content or effectively linkage marker. |
