| In this experiment, three experiments were conducted by vitro and animal tests to study the degradation effects of phytase from different sources on phytate phosphorus in commonly used feedstuffs and application research of laying hens production.In experiment 1: At 37℃,p H5.5.The degradation kineties of phytate phosphorus from corn,soybean, rapeseed meal, cottonseed meal and bran were evaluated at 0,10,20,40,60,120.0,180.0 and 240 min using phytase from Piehia Pastoris and Escherichia coli,respectively. The results indicated that the content of phytate phosphorus in different feedstuffs had large differences(P<0.05);Different enzymes and different feedstuffs have a significant impact on phytate degradation rate(P<0.01); The 240 min degradation rate that phytase B hydrolyzed rapeseed meal was most significantly higher than the phytase A(P<0.01).The 240 min degradation rate that two kinds of phytase hydrolyzed corn, soybean meal, cottonseed meal and bran was not significantly different(P>0.05),but the 240 min degradation rate that phytase B hydrolyzed corn, soybean meal, cottonseed meal and bran was higher than the phytase A.It showed that the overall effect that phytase from Piehia Pastoris hydrolyzed phytate phosphorus in five kinds of commonly used feedstuffs was better than phytase from Escherichia coli.In experiment 2:Animal metabolism experiments were conducted to further observe the apparent digestibility of nutrients of phytase from Piehia Pastoris and Escherichia coli on nutrients in corn, soybean, Rapeseed meal, cottonseed meal, Peanut meal feed ingredients,respectively.forty-eight cocks with healthy body and similar weight were randomly divided into four groups of 12 chickens, a total of four times gavage, cocks would have a rest about 10 days after gavage. Each cock was forced fed five kinds of test feedstuffs 50 g alternately. The excretion was collected two days. Test results were as follows:â‘´Nutrient digestibility of corn:The nutrient digestibility of crude protein, crude fat, calcium(Ca), phosphorus(P), phytic P,ash of Phytase A on corn were 51.81%,12.90%,34.59%,44.73%,7.67%,36.32% higher than the control group(P<0.05);The nutrient digestibility of crude protein, crude fat, Ca, P, phytic P,ash of Phytase B on corn were 42.36%,11.58%,27.80%,48.40%,7.08%,24.51% higher than the control group; The nutrient digestibility of crude protein, crude fat, Ca and ash of Phytase A on corn were 16.38%ã€1.49%ã€16.41%ã€15.65% significantly higher than phytase B(P<0.05); The nutrient digestibility of P of Phytase B on corn was is significantly higher than Phytase A(P<0.05). The digestibility of phytic P of corn was no significant differences between Phytase A and B.⑵Nutrient digestibility of soybean meal: The nutrient digestibility of crude protein, crude fat, Ca, P, phytic P,ash of Phytase A on soybean meal were 9.62%ã€32.63%ã€18.09%ã€31.74%ã€14.06%ã€28.21% higher than the control group(P<0.05); The nutrient digestibility of crude protein, crude fat, Ca, P, phytic P,ash of Phytase B on soybean meal were 15.26%ã€34.25%ã€12.31%ã€40.73%ã€15.39%ã€19.72% higher than the control group(P<0.05); The nutrient digestibility of Ca and ash of Phytase A on soybean meal were 10.28%ã€10.58% higher Phytase B(P<0.05); The nutrient digestibility of crude protein, crude fat and P of Phytase B on soybean meal were 10.28%ã€10.58% higher Phytase A(P<0.05); The digestibility of phytic P of soybean meal was no significant differences between Phytase A and B(P>0.05).â‘¶ Nutrient digestibility of rapeseed meal: The nutrient digestibility of crude protein, crude fat, Ca, P, phytic P,ash of Phytase A on rapeseed meal were 11.37%ã€28.41%ã€34.30%ã€26.58%ã€8.39%ã€21.54% higher than the control group(P<0.05); The nutrient digestibility of crude protein, crude fat, Ca, P, phytic P,ash of Phytase B on rapeseed meal were 4.8% 〠19.34% 〠20.70% 〠22.87% 〠7.80% 〠23.55% higher than the control group(P<0.05); The nutrient digestibility of crude protein, crude fat, Ca, P of Phytase A on rapeseed meal were 6.91%ã€11.24%ã€17.15%ã€4.81% higher Phytase B(P<0.05); The digestibility of phytic P and ash of rapeseed meal was no significant differences between Phytase A and B(P>0.05).â‘· Nutrient digestibility of cottonseed meal: The nutrient digestibility of crude protein, crude fat, Ca, P, phytic P,ash of Phytase A on cottonseed meal were 1.32%ã€21.55%ã€34.00%ã€12.20%ã€8.25%ã€25.12% higher than the control group; The nutrient digestibility of crude protein, crude fat, Ca, P, phytic P,ash of Phytase B on cottonseed meal were 5.35% 〠14.19% 〠26.37% 〠12.02% 〠5.35% 〠21.58% higher than the control group(P<0.05); The nutrient digestibility of crude fat, Ca, phytic P,ash of Phytase A on cottonseed meal were 8.57%ã€10.36%ã€3.07%ã€4.51% higher Phytase B(P<0.05); The nutrient digestibility of crude protein of Phytase B on cottonseed meal was 4.08% higher Phytase A(P<0.05); The digestibility of P of cottonseed meal was no significant differences between Phytase A and B(P>0.05).⑸Nutrient digestibility of peanut meal: The nutrient digestibility of crude protein, crude fat, Ca, P, phytic P,ash of Phytase A on peanut meal were7.50%ã€6.66%ã€29.07%ã€16.45%ã€12.52%ã€24.01% higher than the control group(P<0.05); The nutrient digestibility of crude protein, crude fat, Ca, P, phytic P,ash of Phytase B on peanut meal were 5.39%ã€3.54%ã€14.19%ã€13.10%ã€11.77%ã€14.77% higher than the control group(P<0.05); The nutrient digestibility of crude protein, crude fat, Ca, P and ash of Phytase A on peanut meal were 2.23%ã€3.23%ã€17.33%ã€3.86%ã€10.84% higher Phytase B(P<0.05); The digestibility of phytic P of peanut meal was no significant differences between Phytase A and B(P>0.05).In experiment 3: An animal experiment was conducted to evaluate the effect of diets that added two kinds of phytase on growth performance, blood parameters and apparent nutrient digestibility of nutrients in growing chicken. A total of 200 ten weeks growing chicken with healthy body and similar weight and performance were randomly assigned to 5 groups, consisting of 10 pens of 4 birds each.
