In this study, 400 white Liancheng and white gaiya progeny populations were used as experimental animals. The genetic variations of OIH gene and FSHβ gene were detected by DNA Poll technology. These two candidate genes were cloned and sequenced. The relationship of the genetypes including OIH gene and FSHβ with breeding traits were investigated. The results are as follows:1.The partial of Exon10-Exon12 and Intorn10-Intorn12 of OIH gene on duck were cloned. DNA Poll were used to screen the SNP in the coding region and its flanking sequences of duck OIH gene, 4 new SNPs were detected, and c.316 A > G, c.1287 A> C resulted in Acc I and Alu I restriction sites changed, the frequencies of G and A allele of Acc I-RFLP were 0.33, 0.67 respectively, the frequencies of C and A allele were 0.91, 0.09 of Alu I-RFLP polymorphic sites; association analysis showed that there was significant associations between the c.316 A > G site with breeding traits. c.316 A > G was associated with egg production of 420 days(P=0.05) 、 eggshell strength(P <0.05) correlation. Association analysis showed that there were significantly or very significantly associations between the c.1287 A> C site with breeding traits. c.1287 A> C was significantly associated with egg production of 270 and 300 days(P <0.05) correlation,and very significantly associations with egg production of 330,360,390 and 420 days(P <0.01) correlation.2. The partial of the third exon coding region sequence of duck FSHβ gene was augmented by PCR. 7 new SNPs were identified in the FSHβ gene sequences. c.82C>G site and c.308T>G site were detected by Alu I and Ban II. The result showed that, frequencies of C allele and G allele in duck were 0.24 and 0.76 for the Alu I sites, respectively. Frequencies of T and G allele were 0.64, 0.36 respectively for the Ban II-RFLP polymorphic. Association analysis showed that there was significantly or very significantly association between the c.82C>G site with the breeding traits, was associated with first egg weight(P<0.01), Albumen height(P<0.05) correlation. Association analysis showed that there was very significantly association between the c.308T>G site with the breeding traits. c.308T>G site was associated with egg production of 120 days(P <0.01) correlation.3. Hapiotypes inferred based on the two genetic mutations were refactored by PHASE software, association analysis showed that there was significantly association between the Hapiotypes with the late egg production(EP330d,EP360 d,EP390d, EP420d) on OIH gene, and there was significantly association between the Hapiotypes with the early egg production(EP120d,EP180 d,EP210d,EP240d) on FSHβ gene.
