| The Neillia sinensis Oliv. mainly distributed in Shaanxi, Yunnan and other provinces, belonging to Rosaceae Neillia. It is Chinese’s endemic specie which is a very promising wild shrub with a graceful posture, beautiful leaf, light pink flower, red fruit, collecting flower, foliage, fruit as a whole. Flowering from May to June and fruiting from August to September. Within the blade contains sorbitol and tannin, the leaf has a highly medicinal value. Therefore, it has a great development prospect in the aspects of landscaping design and medicinal science. In this study, we eventually want to obtain a certain number of seedlings through tissue culture, protect and use the germplasm resources of Neillia sinensis Oliv., increase the variety of landscape trees and shrubs and abundant the diversity of the urban landscape, then raising the city landscape taste.In this study, stem segments with axillary buds selected as the explants of Neillia sinensis Oliv. Explores the impact of sterilization method for their culture on primary culture stage; the best ratio scheme of proliferation for tissue culture seedlings by different plant growth regulators and concentrations on proliferation culture stage; the influence of different rooting medium and hormones with different types or concentrations on tooting stage; the points for attention on hardening and transplanting process. Therefore, this experiment study from explants disinfection establish the sterile system to rapid propagation in vitro conditions, rooting and acclimation of transplanted, series of studies provide a systematic basis for Neillia sinensis Oliv. tissue culture techniques.Results are as follows:1. The best disinfection treatment for axillary bud stem of Neillia sinensis Oliv. is 75%alcohol 30s+0.1%HgCl2 8min.2. The best concentration of medium and hormone for Neillia sinensis Oliv. tissue culture seedlings proliferation is MS+6-BA 1.0mg/L+NAA 0.2mg/L. The plantlets growth strong and rapid, more clustered shoots, color of the buds are dark green, its multiplication factor up to 4.13 and the height of clustered shoot is 2.97 cm.3. The best basic medium for rooting is 1/4MS and the best hormone type and concentration is IBA 0.7mg/L. The average number of roots is 5.03 and the average length of root is 2.58 cm, the rooting rate is 63.33%. Healthy plant, thick taproot, more fibrous roots and dark green leaves.4. Select the tissue culture seedlings with robust plant growth and height of more than 2cm for hardening and transplanted, select vermiculite: sand: humus(1: 1: 2) as culture medium, routinely culture for 20 d, the survival rate is 36%. |
