Dendrocalamus farinosus somatic mutant No.29 and the plant of seeds(CK) were employeed as the experimental materials. The plant height, diameter at breast height, fresh weight and dry weight of stem, bamboo shoot emerging amount, ash content were investigated to show the phenotypic differences. The cellulose and lignin content, fibre quality, culm transverse sections were determined to show the fibre’s differences. Net photosynthetic rate(Pn), stomatal conductance(Cond), Chlorophyll content and soluble polysaccharide content were determined to show the physiological differences. Meanwhile, RNA high-throughput sequencing were carryed out to elucidated the mutant mechanism. The main results are as follows:In No.29, the plant height, diameter at breast height, fresh weight and dry weight of stem are significantly higher than CK. The ratio of dry weight and fresh weight in No.29 and CK are 21.42%, 21.83%, respectively. The cellulose content in the mutant No.29 increased 12.89%, compared with CK. The ash content, curl index and kink index is significantly lower than that of CK. The shooting number, lignin content and fiber fines content are not significant compared with CK. The plant biomass of No. 29 overall is superior to the control. It has good biological characteristics.Net photosynthetic rate(Pn) and stomatal conductance(Cond) have no difference than CK while the Chlorophyll content and soluble polysaccharide content significantly higher than CK. The soluble polysaccharide content is higher for 31%. The content of chlorophyll and soluble polysaccharide in No. 29 is significantly higher than control. It has excellent physical properties.A total of 48771 ALL-Unigenes in the 68800 ones were annotated in the database of COG, GO, KEGG, Swiss-Prot, TrEMBL and Nr. 2,119 Unigenes contained SSRs.CesA, SUS, PAL, 4CL, CAD, LAC, prx, MYB, NAC, WRKY, bZIP, which are key genes durring the growth of Dendrocalamus farinosus, are analysised by heatmap. The expression of gene of CesA, SuSy, UDPase, transcriptional factor MYB in the mutant No.29 was up regulated by 117%, 205%, 115%, 214%, respectively. The kinesin was up regulated by 10 fold while the dynein was up regulated by 760%. The results indicated that the significant changes of cellulose content and fiber morphology have the genetic bases in the mutant No.29.
