| Thinopyrum intermedium(2 n = 6 x = 42, JJJSJSStSt) is an important source of genetic variability for improving cultivated wheat,which has developed root system, strong resistance to harsh environment and grow prolifically. It can resist many kinds of diseases which like the powdery mildew, the rust,the yellow dwarf and so on. Now,we have selected 36 germplasm Lines, which were developed from progenies between Th. intermedium and Triticum aestivum cv. Yannong15. In the present study, they were identified and analysed by methods of morphology, cytology and molecular biology. The main results were as follows:1. Field investigation results show that the heading date and flowering date of 36 germplasm lines are 10 days later than yannong 15. The height of all plants are between Th. intermedium and Triticum aestivum cv. Yannong15. All germplasm Lines grow strong, lush and strong resistance to diseases. The cytological identification shows that there were 42 chromosomes in 19 germplasm lines, among the 19 germplasm lines, 4 germplasm lines’ chromosome configuration are 21 II, which can be alien substitution line, translocation line and intogression line. Among the other germplasm lines, 15 germplasm lines’ chromosomes in RTCs are 44, there are 3 germplasm lines which chromosome configuration are 22 II, we can infer that they are disomic alien addition lines. 2 germplasm lines’ chromosomes in RTCs are 43, which can be monosomic addition line.2. The resistance identification showed that the ‘20120211’ was nearly immune to powdery mildew of adult in field whatever seeding in lab. Compared with the ‘Shanrong 3’ which was highly resistant to salt, to identify the salt resistance of ‘20120211’. The result shows that the ‘20120211’ is a new germplasm material which salt resistance is higher than ‘Yannong 15’. The results of GISH showed that there were two chromosomes of ‘20120211’ with obvious hybridization signals, which further proved that ‘20120211’ was an alien disomic addition line with a pair of Thinopyrum intermedium Ee chromosomes in wheat background. Using 440 pairs chromosome specific molecular markers of group Ee,430 pairs chromosome specific molecular markers of group Eb, and 430 pairs chromosome specific molecular markers of group St to analyze the ‘20120211’, the results shows that the marker BE494262 could reveal the specific band of Thinopyrum intermedium and be used to identify ‘20120211’. Using Elytrigia elongata and the addition line of Cs-Elytrigia elongata(1Ee-7Ee) to further located the BE494262 at 2Ee, which identified that the added chromosomes of ‘20120211’ came from the 2Ee of Thinopyrum intermedium.3. The resistance identification showed that the ‘20120242’ were nearly immune to powdery mildew of adult in field whatever seeding in lab, and it highly resistance to stripe rust. The results of GISH showed that there were no chromosomes of ‘20120242’ with hybridization signals. The results of FISH showed that there was no signals the same. Becausee the agronomic traits of ‘20120242’ is obviously different to ‘Yannong 15’, so we can infer that the ‘20120242’ is a intogression line. Using 440 pairs chromosome specific molecular markers of group Ee,430 pairs chromosome specific molecular markers of group Eb, and 430 pairs chromosome specific molecular markers of group St to analyze the ‘20120242’, the results were as follows: the marker BF2007042, Xpsp3113 and Xwmc74 could reveal the specific band, it showed that there were a small amount of genetic material which come from Ee, Eb, St in the ‘20120242’.The three markers can be used to identify it. Because of the Yannong15 was high-susceptible to powdery mildew and stripe rust, so we can infer that the ‘20120242’s resistances of powdery mildew and stripe rust were come from the Thinopyrum intermedium. |
