Chimonanthus praecox is, one of the Chinese top 10 famous special-smell flowers, however, the composition of floral emission were proved to be varied greatly among different varieties,after GC-MS analysis of several genotypes of full-open flowers. Linalool was always been detected as one of the main compound that make great contribution to flower scent. The promoter of linalool synthase(LINS) of 2 genotypes H29 and H64 were cloned and the contrast of expression of LINS as well as linalool substances emissions were investigated.Moreover, the expression vector of Cp LISP-H64 deletion was constructed by the 5’promoter deletion method. The expression of deletion promoter was verified using Arabidopsis protoplasts. The main contents and results in this study can be summarized as follows:1. 86 kinds of compounds were obtained 5 genotypes of Chimonanthus praecox.The volatile amount of H93 was significantly higher than the other genotypes and took linalool and benzyl ester as its main compounds. Volatile amount of H64 came the second which has the same component as H93.The linalool and β-ocimene were the main volatiles in the H36.Different from the other genotypes,β-ocimene was the main. volatile in SW001 while there was little linalool has been detected.2. Regarding the particularity of volatile composition of SW001, we analyzed the floral scent in different stages of SW001 including bud, display-petals, initiation, full-open and senescence. 60 compounds were found in SW001. β-ocimene. 1-caryophyllene.Methylis salicylas.and benzyl alcohol were the main compound. Linalool and benzyl were absent in all the stages of flowers.And it showed that the content of main volatile rised slowly from bud period to infant period and reached its peak at the blooming period.declined at the end of flowering.3. The release of linalool in H29 and H64 were identical with expression level of its synthetase. By Cloning and comparing the two linalool synthase promoter with the length of 1106 bp, only 3 single nucleotide differences were found which indicates that the release of linalool different level. may not related to the different promoter structure.4. Light, hormone and cis elements related to abiotic stress were found by Analysis of Cp LISP-H64 structure. Using 5 ’promoter deletion method, the expression of deletion promoter was verified using Arabidopsis protoplasts. The deletion of the translation initiation site of 750bp-1006 bp showed the highest activity to be considered the main functional areas.
