Guanylate-binding protein family, member 6(GBP6) in pig belong to the four major families of large Guanosine Triphosphatase (GTPases) which contribute to interferon (IFN) responses in a variety of organisms. Several SNPs were identified in the coding region of porcine GBP6, one of which changed the start codon for one alternative splicing transcript of GBP6. So we suppose this SNP may have a potential influence to the function of porcine GBP6, therefore could be a good functional candidate for immune-response related traits. Therefore we analyzed the proportion of SNPs in porcine GBPs and the effect of disease-causing missense mutations arising from the pGbp6.Semi-quantitative RT-PCR revealed that pGBP6was highly expressed in immune-related tissues, spleen and lung. Single nucleotide polymorphism (SNP) in coding region of pGBP6 was detected in different breeds of pig, Duroc, Landrace, Luchuan and Dongshan by Nlaâ…¢ restriction enzyme digestion and the result shows that different population differs greatly in the genotype and allele frequencies.We performed association analysis for 22 hematological traits in an F2 DurocxErhualian resource population (n=306). Three main components,11 leukocyte traits,6 erythrocyte traits and 5 platelet traits were measured after treatments with mimic viral infection Polyriboinosinic-Polyribocytidylic acid (polyI:C). Different genotypes of this SNP had significant associations with 5 hematological traits, including Platelet count (PLT) (P<0.01), Plateletcrit (PCT) (P<0.01), White blood cell (WBC) (P<0.01), Neutrophil count (NE) (P<0.05) and Red blood cell count (RBC) (P<0.05) through a linear mixed model. Genotype-Phenotype correlation analysis shows the homozygous and heterozygous genotype classes have different phenotypic distribution. Our results confirmed that the GBP6 gene might have important roles in porcine immune responses, and these results provide useful information for further studies.
