Application of molecular genetic approach in the pig can help to understand gene structures, functions, and its relationship with phenotypes and traits. It should lead to the development of the theory on animal breeding, and supply more new approaches and more new candidate genes for marker assisted selection to the pig breeding.In this study, SNPs in the Exon4 of porcine SDHD, the RH mapping, the association between the SNPs genotypes and the traits in the Tongcheng porcine group, and the expression profile on SDHD gene were investigated. The main results of the research are:1. To clone the segments from the Exon4 in SDHD gene, cDNA sequence from our 55-day fetal skeleton muscle cDNA library was used to design the specific primers;2. After the detection of SSCP, PCR products which respresent the two type of homozygotes weresequenced. Two SNPs have been found, one of the two mutations caused amino acid change but no restriction enzyme was found to recongnize this mutation, the other doesn't cause amino change but it can be detected by the Restriction endonuclease Mbol;3. Allele frequency and significant test of the alleles distribution among the different nonrelatived pig groups were conducted by the detection of SSCP and Mbol PCR-RFLP. The results showed that the significance of the frequencies distribution of alleles and genotypes is different.4. Association study between production traits and Mbol-RFLP in SDHD genes showed that the relationship between the SDHD gene and eye muscle area is very significant (P=0.0067), while there is nearly significant relationship between the SDHD gene and the marbling score (P=0.0614).5.
