Objective:Through established and compared the advantages and disadvantages of three kinds of canine cataract models, choose the better model as continuous test. And examined the relationship between the cataract development process and the AQPO expression amount, to explore the influence of AQPO to cataract formation.Methods:(1) Choose 60 local hybrid dogs, which about six mouth, divided into three groups randomly, namely Fenton cataract model group, STZ model group, selenite model group and the control group. Observed by slit lamp and photographed daily, according to the LOCSⅢ grading and classification standards recorded the structure changes of lens. (2) Selected 40 local hybrid dogs were randomly divided into two groups, the model group and control group, recorded the cataract formation process according to the LOCSⅢ grading standards, and the time of each node in the model group level detection formed, measured the AQPO expression of lens in each classification by RT-PCR.Results:(1) Selenium model cataract, diabetic cataract model and hierarchical model of oxidative damage in cataract formation time were C1 (d):1 ± 0.32,30 ± 2.0 and 7 ± 0.8. C2 (d) were 5 ± 0.36,60 ± 2.5 and 10 ± 0.68. C3 (d) were 11 ± 0.48,150 ± 6.25 and 15 ± 1.0. C4 (d) was 0,180 ± 6.5 and 24 ± 1.54. C5 (d) were 0,240 ± 12 and 48 ± 1.52. Selenium cataract model had the shortest time, but highly toxic, up to 100% mortality rate. Oxidative damage model shorter than the diabetic cataract molded. (2) The number of dogs in C1 of selenium cataract formation, diabetic cataract model and oxidative damage cataract model were 10,7 and 10, and at C2 were 10,4 and 10, when the C3 formation were 0,3 and 10. When the C4 were 0,3 and 10, when the C5 were 0,3 and 10. And the three control groups of cataract model were all 0. Therefore, the stability of oxidative damage model batter than the diabetic cataract model and selenium model. (3) The three kinds of modeling methods all have operability, oxidative damage model compared with diabetic cataract and selenite was slightly difficult. (4) The results of the slit lamp observed:Selenium model:Id after injected cataract selenium selenite saline the dogs eyelid substance had some punctate opacities, in 5d the punctate opacities deepen and linked as mydriasis, cortex and surrounding oval shadow were visible, a large number of vacuoles, after 11d the shaded area expanded to form a sheet, the amount of vacuoles increased, the details shaded areas fundus were not clear. After 30d injection of STZ diabetic cataract lens of a small number of small vesicles, after 60d the vesicles increased in groups, in 150d the vesicles even after a patchy shadows, surrounded by extended to the equator, retinal shadow details are not clear, skin-like shadows deepen, afterl80d expand the scope of the shadow of fundus details are not visible, after 240 d lens cortex and nuclear quality completely ashen white opacity. Injection of Fenton oxidation cataract after 7 d formation of lens cortex dotted white dots shadow but observable fundus oculi, white point 10d after injection lens cortex punctate distribution increases gradually deepen a white shadow sectors distributed, injection after the sector distribution 15d lens cortex white shadows deepened, the range continues to expand to form a schistose,24d after injection of flake white shadow extended to the whole lens cortex,48d after injection of mature cortical cataract formation.(5) Oxidative damage cataract model development process AQP0 expression data processing using the amount of change relative quantification △△Ct method, according to the standard curve of each gene, by testing samples of the CT data corresponding to the data, this comparison Fenton oxidation injection cataract formation after 7d、10d、5d、24d、 30d、40d、48d of the expression level of AQPO. AQPO expression quantity in 7 d for 1.90 times in the control group, the expression level at this time for the entire detection peak value, AQPO expression levels after 7d showing a decreasing trend, the first lOdays was 1.83 times,15 days was 1.5 times,24 days 1.1 times, first for a minimum of 0.6 times 48d. The distribution is consistent with the degree of lens opacity of AQPO.Conclusion:(1) Experimental study on oxidative damage model is suitable for cataract cataracts. (2)There is a close relationship between the incidence of AQPO and Oxidative damage cataracts.
