In order to study the molecular mechanisms of yak about altitude hypoxia adaptation, Inthis study, four yak breeds (including Gannan,Datong,Tianzhu and Pali) in high altitude wereselected as experimental animals, three genes MMP3, ADAM17and ARG2exon region wereamplified using molecular biology techniques, The SNPs were detected by using mixed DNApool sequencing technology combined with High resolution melting(HRM)technique. Finally,using statistical analysis software of SHEsis, so as to find correlations between the candidategenes SNPs and the adaptability of the plateau of yak. In order to filter out the molecularbeacons concerning the adaptation to high altitude hypoxia environment, so as to provide atheoretical basis for the further study of yak high altitude hypoxia adaptation mechanisms.The main results are as follows:2specific SNPs (rs2977G'A and rs4331C'G) were detected in yak’s MMP3gene.The results of HRM genotyping showed that2Dominant genotypes GG and CC weredetected; For four yak haplotype analysis, haplotype GC yak population was significantlyhigher than in other haplotype. Suggested that haplotype GC can adapt to hypoxia, yakMMP3gene can be used as molecular beacons adapt to high altitude hypoxia environment.2specific SNPs (rs21928G'A and rs38991A'G) were detected in yak’s ADAM17gene. The results of HRM genotyping showed that2Dominant genotypes GG and AA weredetected; Haplotype analysis found that high-altitude Pali and with1/2yak blood Datongwere significantly different with low high-altitude Tianzhu. Presumed advantage haplotypeGA can adapt to hypoxia, Yak ADAM17gene can be used as molecular beacons adapt to highaltitude hypoxia environment.2specific SNPs (rs30979C'A and rs32860T'G) were detected in yak’s ARG2gene.The results of HRM genotyping showed that rs30979(C'A)mutation,high altitude Pali,Gannan and Datong with low-altitude Tianzhu have significant differences between genotypefrequencies or Allele frequencies, and other breeds were not significant. Suggested that (C'A)mutation may be conducive to yak to adapt to hypoxic environment. Haplotype Pearson’s pvalue than the results display, in middle altitude Gannan and Datong yak with low altitudeTianzhu yak were significant differences (P <0.01). Presumed advantage haplotype CT may be more adapted to hypoxia, yak ARG2gene can be used as molecular beacons adapt to highaltitude hypoxia environment.
