To study the histological structures of the3,40,50,60,72,86,95and165daypost-partum Chang Bai pig ovaries, the features of the development of all follicles, thedistribution of mucopolysaccharide and the location of H1foo, we used common paraffinsectioning method stained with HE method, PAS method, immunohistochemistry andimmunofluorescence methods.HE reaction showed that the following histological features: the developmental processof pig ovary was divided into three periods----proliferation of oogonia (P1,3rd daypost-partum), slow growth of follicles (P2,40th to60th day post-partum) and rapid growth offollicles (P3,72nd to165th day post-partum).In P1, boundary between cortex and medullawas dim, there was rete ovarii in the ovary, and oogonia proliferated significantly. The densecluster of oogonia, nested oogonia groups and synplasma-like cell groups made up75%of thewhole cortex. At the same time, a number of degraded oogonia were observed. In p2, thecortex became thinner and medulla thickened with age, and the rete ovarii had alreadydegraded. Moreover, the number of primordial follicles and primary follicles increasedgradually, and the primordial follicles reached its maximum until60day post-partum pigovary. In the meanwhile, a vast number of degraded primordial follicles were observed. In P3,the pig ovary further increased, and boundary between cortex and medulla was obvious. Thenumber of growing follicles increased gradually. Compared to the former two periods,primary follicles reached its maximum in86day post-partum pig ovary and secondaryfollicles reached the maximum in72day post-partum pig ovary. Until95day post-partum pigovary, tertiary follicles appeared.PAS reaction showed that the mucopolysaccharide was mainly located in the stromaaround the oogonia and follicles, the envelope of nested oogonia and syncytium-like cells, theenvelope of the oogonia, the envelop of the growing follicles and the zona pellucid, thefollicular fluid in secondary follicles and tertiary follicles, tunica albuginea, envelope ofcorpus luteum, connective tissue, blood vessel wall and rete ovarii.H1foo was detected in the cytoplasm of the oogonia and atrestic oogonia, ooplasm offollicles and atrestic follicles, granular cells of follicles. And weak positive reaction was detected in the nucleus of oogonia and oocytes. The positive intensity showed an upwardtrend with the development of follicles.In conclusion, in the pig ovaries, H1foo is involved in the growth and degradation ofoogonia and follicles, and the development and mature of the oocytes.
