| Prunus mira koehne Kov et. Kpst with lots of good characteristics, is a kind of special germplasm resource in the Tibet Plateau of China. However, little attention has been devoted to systematic study of Prunus mira koehne Kov et. Kpst due to the limitation of geography. In this study, with wild Prunus mira koehne Kov et. Kpst from the Tibet Plateau and other peach species came from Zhengzhou and Harbin, two molecular markers (SSRs and ISSRs) have been used to analyze:i characteristics of genetic diversity among wild Prunus mira koehne Kov et. Kpst accessions, ii genetic relationships between wild Prunus mira koehne Kov et. Kpst accessions and other peach species. The aim of the study was to offer theoretical guidance to conserve and utilization Prunus mira koehne Kov et. Kpst germplasm resources efficiently. Results as follows:I SSR Polymorphism:The genetic diversity in72peach accessions were assessed using10SSR primer pairs. A total of55alleles were detected and the percentage of polymorphic alleles (PPA) were100%; The mean value of effective number of alleles (Ne) was1.608; The mean value of Nei’s gene diversity index (H) was0.342; The mean value of genetic diveristy within populaitons (Hs) was0.216; The mean value of proportion of species genetic diversitiy attribted to among populatin variation (Gst) was0.329; The mean value of gene flow from Gst (Nm) was3.293; The mean value of Shannon’s Information index (I) was0.507. This result suggested that a lot of information of genetic diversity was got by SSR markers. The result of UPGMA and PcoA indicated that the altitude, latitude and longitude have a significant impact on genetic diversity, and influence the genetic variation of46wild Prunus mira koehne Kov et. Kpst accessions.72accessions were clusted into two main groups, where most of wild Prunus mira koehne Kov et. Kpst were in one group, and most of other varieties were in another one. Some of wild Prunus mira koehne Kov et. Kpst accessions had raltionship with southern peaches from Henan Province.II The result of genetic diversity by ISSR:The genetic diversity in72peach accessions were assessed using10ISSR primer pairs. A total of67alleles were detected and the percentage of polymorphic alleles (PPA) were77.12%; The mean value of number of alleles (Na) was1.7712; The mean value of effective number of alleles (Ne) was1.3817; The mean value of Nei’s gene diversity index (H) was0.2318; The mean value of Shannon’s Information index (I) was0.3565; The mean value of genetic diveristy within populaitons (Hs) was0.2319; The mean value of proportion of species genetic diversitiy attribted to among populatin variation (Gst) was0.2307; The mean value of gene flow from Gst (Nm) was1.6678. The result of UPGMA and PcoA suggested that72accessions were clusted into two main groups. 53accessions were in one cluster, including34wild Prunus mira koehne Kov et. Kpst accessions and19other peach species;19accessions were in another one, including12wild Prunus mira koehne Kov et. Kpst accessions and7other peach species. And the result indicated that the geographic location has litttle impact on genetic relationships, and wild Prunus mira koehne Kov et. Kpst accessions showed some raltionship with southern peaches from Henan Province.The total results by ISSR and SSR as follows:the geographic location has impact on genetic relationships in72accessions; wild Prunus mira koehne Kov et. Kpst accessions showed some raltionship with southern peaches from Henan Province; SSR has high polymorphism and more stable than ISSR in this study. |
