Stripe rust, caused by Puccinia striiformis f. sp. tritici, is one of the most importantdiseases of wheat in China as well as in the world. Wheat stripe rust epidemic areas inLongnan Gansu, could be classified different popular small area according to the complicatedterrain conditions. Studying on propagation of stripe rust in the small area through SCARmethod, by extension, we could deduce the propagation of wheat stripe rust in the othersimilar regions. It provides great strategies and methods for comprehensive prevention ofstripe rust by revealing epidemic characteristics.Three different elevation test points (A, B and C)were selected and inoculated with racesCYR33, CYR17and CYR32, separately, at dayside of the valley with gap of about200meters in mutability area of Longnan Gansu. Genome DNA of test point leaves wereextracted and amplified by SCAR method to analyze the frequency of different physiologicalraces and to deduce the propagation of wheat stripe rust in small regions. The main resultswere summarized as follows:(1) SCAR markers of CYR33and CYR17were established. SCAR primers of CYR33were designated S261F33/S261R33and S300F33/S300R33while that of CYR17was namedS13F17/S13R17.(2) Wheat stripe rust could spread in different altitude of Longnan Gansu to be infectionsource of spring epidemic after overwinter or seedlings of autumn. The transmissionefficiency of stripe rust was related to spatial distance that to be more effective and efficientwith a closed distance.
