Hemerocallis citrina Baroni is perennial herbaceous plant of the Lily family and traditional anthophagous vegetable in China. There was a long cultivation historical standing and extensive distributed areas of Hemerocallis citrina Baroni, but there was scarcely professional organizations to collect and sort out resources of Hemerocallis citrina Baroni, so the resources was unclear. Besides, Hemerocallis citrina Baroni has colchicine and can cause poisoning easily. Traditional processing was complicated and reduces nutritions with high costs. This study investigated and collected different cultivars of Hemerocallis citrina Baroni in China, established germplasm resources nursery, used solvent extraction-high performance liquid chromatography to compare colchicine content of different cultivars, and identified genetic relationships between seventeen kinds of Hemerocallis citrina Baroni using SRAP molecular marker, so that provid the basis for innovation and utilization of resources, help resolve toxicity problem of the Hemerocallis citrina BaroniMajor research results:1. Investigated the major producing unit in China, collected29varieties of Hemerocallis citrina Baroni (4wild varieties and6local varieties among that),1118plants in total, and established germplasm resources nursery in Shanxi Agricultural University.2. Optimized extraction process of colchicine. The best colchicine solvent extraction condition with orthogonal test:absolute ethyl alcohol as extractant, Solid-liquid ratio is1:8, constant temperature extraction8h in85℃. Using solvent extraction-high performance liquid chromatography to compare colchicine content of15varieties Hemerocallis citrina Baroni, the result showed that the Changzuizi variety has the highest concentration is13.53μg/g, the Qiaotou variety has the lowest concentration is1.69μg/g.3. Established the PCR reaction system of Hemerocallis citrina Baroni SRAP molecular marker with orthogonal test, analyze17Hemerocallis citrina Baroni varieties with15pairs of primers, get146bands in total,72polymorphic bands among that. It showed that17varieties have rich genetic diversity, SRAP finger-print cluster analysis classify the samples into2kinds and4groups.The study provid the basis for resources utilization, botanical research, species classification and breeding of new variety. Meanwhile, help the genetic map construction and Marker-assisted breeding of Hemerocallis citrina Baroni, so that obtain the new variety has low or no colchicine.
