The experiments study the correlation between the pigment gland size, destiny and gossypol, observing and analysis from Microscopic and ultrastructural aspects during mature and germination stage, using three kind of Near Isogenic Lines cotton but different pigment gland trait, The main results were as follows:1、The pigment gland morphogenesis in cotton seeds. The pigment gland builds in cotton with glands. Stage one, the stage of pigment gland original, which appeared in25days after flowering. The stage tow, the pigment gland dissolution and become a cavity, meddle cell broken up, then become gland cavity, which become more larger day by day in40days after flowering. Stage three, the mature of pigment gland, the gland cavity mature after36h of cotton seeds germination. The Glandless seed and Glands plants near-isogenic lines morphogenesis in cotton. Stage one, the stage of pigment gland original, which appeared around26days after flowering, then maintain to the mature of cotton seeds. Stage two, the pigment gland dissolution and become cavity, after10hours of seed germination. Stage three, mature of pigment gland. The glands mature after72hours of seeds germination. We have not found pigment gland morphogenesis in glandless cotton.2、we analyses the correlation between pigment gland and gossypol. The pigment glands begin to develop during the mature of embryo. We analyses the correlation between pigment gland size and gossypol contain. Results show that it’s positive correlation in pigment gland size, number and gossypol contain when a=0.05. There is no correlation, between gland number and gossypol contain. There have significant correlation between the size and destiny of pigment gland and gossypol contain in cotton with gland and seed glandless cotton when α=0.01.3、We studied the dynamic change of gossypol synthesis and accumulation. The gossypol can’t precipitate when seed embryo mature but appear a little, after membrane lysing. It becomes more in the stage of the pigment gland cavity. When the seed germination, the pigment gland mature, membrane structure larger, other area precipitate much more gossypol and electronic transparent higher. Glandless seed and Glands plants near-isogenic lines in embryo maturation period.Gossypol cannot precipitate during embryo mature stage, gossypol contain is low. In the seed germination period, pigment glands primordium cells have membranous structure, Gossypol cannot precipitate during embryo mature stage, the pigment gland mature, gossypol deposition appear around membranous structures, Then pigment glands cavity increases, gossypol deposition appeared in the membranous structures, Membranous structures adjacent to the fusion. the pigment gland mature, more gossypol deposition appear in the membranous structures, the same as The pigment glands near-isogenic lines.
