| 20lines derived from the progeny of hybrids between wheat-rye substitution lines5R/5AX1R/1Dã€6R/6AX1R/1D and wheat-rye translocation line with substitution line1R/1D X Kezhenã€6R/6A X Kezhen, were identified and analyzed by the methods ofmorphology, cytology, SSR and GISH analysis, which aimed at breeding the smallfragment translocation lines with R group fragment of rye chromosome as well as witha big spike, multi-spikelet,strong resistance and other excellent traits.Morphological observation showed that the plant possess the character of rye suchas big spike, multi-spikelet,strong resistance and so on.The height of plant was mostlyat the range of70cm to90cm,with a strong stem and lodging resistance was strong. Thetiller number was mostly at the range of3to7,and the panicle length was mostly over10cm. The spike shape mainly came out to be three kinds: spindle-shaped, cone-shapedand oblong-shaped. The seeds had both common wheat and rye character, the shape waslong oval and the kernel plumpness was good. The identification results of cytologyshowed the chromosome number of root tip was42at mitosis metaphase and21bivalents were formed at the meiosis metaphase of PMC.The molecular test results showed that20materials amplified with rye specificprimer PSC119.1were contained ingredients of rye chromosome. The test result ofspecific primer SCM2-6RL showed the ingredients of rye chromosome had beensuccessfully inserted into lines2-18,2-19,2-20,2-21,2-22,2-24,2-25,2-26,2-29,2-31,3-1and3-2. The result of primer SCM180-6RL showed the ingredients of rye chromosomehad been inserted into lines2-18,2-19,2-20,2-22,2-23,2-24and2-26successfully. Theresult of primer Xgwm247-6RL showed the ingredients of rye chromosome had beeninserted into lines2-18,2-19,2-21,2-22,2-23,2-24,2-25,2-26,2-29,3-4and3-5successfully. The result of primer SCM40-7R showed the ingredients of ryechromosome had been inserted into lines2-21,2-23and2-25successfully. The result ofprimer SCM206-3RS showed the ingredients of rye chromosome had been insertedinto lines2-18,2-21,2-22,2-23,2-24,2-25,2-26,2-27and2-28successfully. The GISH test results showed that line2-18,2-19,2-20,2-26,2-27,2-31,3-1,3-3and3-5were displayed obviously red fluorescence signal at the end of chromosome andnear the end of chromosome in20lines.Therefore, based on the above test results, the detected20progenies of hybridbetween substitution lines and substitution lines with translocation lines were smallfragments translocation lines, which provide a good foundation material for the wheatgenetics and breeding. |
