Pre-harvest sprouting (PHS) is a serious problem, reduce not only the wheat seeds, but also decrease the quality of flour and end-use application, and causes serious economic losses. Three Pre-harvest sprouting tolerance-associated markers VplA3、 Vp1B3and TaDFRBbF/R, were used to screen232recombinat inbred lines of Longmail5×Longfumail2, aimed at selecting genotype with higher PHS tolerance.Two kinds of alleles were amplified with STS marker VplA3. Longmail5was allele of Vp-lAin, and Longfumail2was allele of Vp-lAhn. In their offsprings,133were Vp-lAin, with an average GI value of0.146968;117were allele of Vp-lAhn with an average GI value of0.13697.Two kinds of allele were amplified with other STS marker Vp1B3. Longmai15was allele of Vp-1Ba and Longfumai12was genotype of Vp-1Bc. In their offsprings,133were allele of Vp-1Ba with an average GI value of0.143491,97were allele of Vp-1Bc with an average GI value of0.130281.For the third marker TaDFRBbF/R, Longmai15did not give the PCR fragment but Longfumai12. In their offsprings,159gave the PCR fragment with an average GI value of0.137758,71did not give the PCR fragment with an average GI value of0.146726.At last,29RIL were selected, which had higher PHS tolerance, by screening result with the three markers and GI value, the number of them are1570、1581、1584、1624、1642、1652、1653、1675、1676、1683、1704、1707、1709、1710、1711、1714、1722、1723、1725Mang、1757、1762Mang、1778、1779、1781、1782、1785、1786、1788and1791.
