Tomatoes are now one of the world’s cultivated a very wide range of vegetable crops. There are more and more study on tomato’s quality traits with people’s quality of life is ever increasing. Soluble sugar content has an important impact on tomatoes’quality triaits. Cultivate high-sugar tomatoes can meet market demand and low-sugar tomatoes can suitable special demand of diabetes. Research on tomato fruits sugar content become more important. On the one hand, the positioning of the soluble sugar content of tomato genes can tomato quality breeding future play a guiding role. It can also lay a foundation for future fine mapping genes of tomato soluble sugar, map-based cloning.The experiment using six generations to analysis the inheritance of tomatoes’soluble sugar content.We use AFLP and SSR to screened out the gene which is closely linked with tomatoes’soluble sugar.1、We choose high-sugar variety12680as female parent and low-sugar variety12574as male parent. Bulid six generations:P1、P2、F1、F2、BC1p1、BC1P2, and test their sugar content. Experimental results show that12680soluble sugar content was6.65%,12574soluble sugar content was1.06%. F2generation of soluble sugar averages3.10%, the highest value is6.85%, it showed forward heterobeltiosis. The lowest value is0.58%, it showed negative heterobeltiosis. F2generation’s sugar content showed continuous change, the gene which is contol soluble sugar content can be considered as quantitative traits.2、The experiment totally chooses256paris of AFLP molecule markers,185pans of SSR molecule markers. Finally24paris of AFLP molecule markers and24paris of SSR molecule markers have favorable polymorphism. F2generation were analyzed for patterns to construct the six linkage groups. It contain51molecule markers, the total length of1421.28cM. There are two markers detected the LOD scores greater than2.5, the distance between Qs2and M12E14is5.58cM, genetic contribution rate is29.18%. The distance between Qs1and M12E14is14.28cM, genetic contribution rate is18.44%.3%Using the molecule marker M12E14to screen50tomato germplasm resources, combining the measuring physiological indicators and marker method, both the identification results coincide. It showed the molecule marker which we get in the experiment is reliable. It can supply excellent tomato germplasm resources for breeding work in the long run.
