The tomato was grown widely all over the world, and it was a kind of important vegetable in china.In recent years, with the development of market economy.the increasing of consumers,the requirements of the tomato’s quality are increasing.In the experiment,molecular marker technique was applied to investigate the fruit smooth gene (F) at the DNA level. Screening AFLP and SSR molecule markers linked to the gene F,and the marker was used to screen tomato plants containing the F gene.The main results of this study was as following:1.In this study tomato contianing F gene12509,without F gene12510and their hybridizing F1,F2progenies were used as the separating groups.using field phenotypic characterization, established smooth fruit and fasciated gene pools.The results indicated that all of F1groups were smooth.actually segregation ratio of F2groups was3.174:l,suggesting that the F gene was a single dominant gene.2.Screened out seven markers which tightly linked to F gene of tomato,among them there were six AFLP markers E13M08,E11M07-B,E11M12-D,E11M08-A,EO1M10and E10M16-E,linkage distance was6.4,8.5,10.9,17.1,23.5and24.2cM;one SSR marker TEI0069,linkage distance was12.1cM.3.Application the obtained AFLP marker E13M08,used70germplasm resources as experimental material to detect the AFLP markers.the inosculation rate is up to90%.this results make a foundation for molecular breeding and gene cloning. |
