The research used the main cultivars of cassava SC124and SC205as the experimental material for the study on the following aspects:it established the SC205tissue culture’s rapid propagation system by taking SC205stem with axillary buds as the explants and discussed the factors that influence the rapid propagation of cassava’s tissue cultures; It explored the factors that affect the regeneration of cassava by taking the lamina, stem and petiole of SC124and SC205as the experimental material, which aimed at providing theoretical and technical foundation for the rapid propagation of cassava’s isolated pieces and the genetic improvement through genetic engineering technology. The main research results are as follows:1. The third to sixth knob of cassava’s terminal bud is suitable for explants, this part is easier to sterilize, so the explants’rate of survival is relatively higher; soaked the explants with0.1%carbendazim solution for one-hour pretreatment can reduce the contamination rate; The appropriate sterilization method for the explants sampled on July to August is75%ethyl alcohol for20seconds+0.1%HgCl2for10min; The appropriate sterilization method for the explants sampled on September to October is75%alcohol20s+0.1%HgCl2for12min; The most appropriate sterilization method for hydroponic buds is process5min in0.1%HgCl2solution.2. The suitable primary culture medium for cassava’s sprouting of axillary bud is MS+6-BA0.5mg/L, the suitable medium of subculture proliferation is MS+6-BA0.5mg/L+KT0.5mg/L and it proliferates in the form of bud, the growth coefficient reaches4.5. The relatively suitable rooting medium is1/2MS+NAA0.1mg/L, the rooting rate is100%and the average root number is5.1.3. In the experiment of using two varieties of lamina, petiole and tender stem segments to induce adventitious buds, it is found that SC124could induce callus from lamina, petiole and tender stem segments and differentiate adventitious buds, which could produce regenerated plantlet while the lamina can directly differentiate adventitious buds; for SC205, only its lamina can differentiate adventitious buds by inducing callus, while the callus induced by petiole and stem segments can not differentiate adventitious buds.
