| The popularization of banana improved variety is one of the major factors to improve the planting benefit and to promote the development of the banana industry. The successful application of tissue culture techniques in breeding of bananas has given an enormous impetus to the rapid breeding of banana healthy seedling. Tissue culture techniques currently applied in the banana seedling breeding mainly use semi-solid medium, which makes low multiplication coefficient, intensive labor, high production cost and low mechanization and automation. There were three different of liquid cultivations used in this test, which were "Double Bottle Integration" Temporary Immersion Culture System, Rotary Liquid Culture system and static liquid culture, in order to research the banana’s micro-propagation and a new way of cultivation that could result in a high multiplication coefficient, high quality of tissue culture plantlets and low production cost. Meanwhile, the introduction of new bred varieties of banana "Hongyan1" in Guangxi was aimed at confirming its adaptability and productivity in Nanning and Hepu. The main results were summarized as follows:1. When making the tissue culture of banana through Rotary Liquid Culture system and Temporary Immersion Culture system, the multiplication coefficient was higher. They were5.90and4.57respectively, which were2.00and1.56times respectively in the traditional semi-solid culture. The Rotary Liquid Culture system had a obvious advantage on multiplication coefficient, stem diameter and root length, while "Double Bottle Integration" Temporary Immersion Culture system was the best in the plant height and root numberproliferation rate. These three ways all brought a lower browning degree than Semi-solid culture.2. Immersion frequency of5min/2h was the best in the banana tissue culture tested by Temporary Immersion Culture system.3. In the banana tissue culture test of Rotary Liquid Culture system, the best6-BA concentration for the growth of the tissue culture planets was3.0~4.0mg/1. The best inoculation density in all respects was20ml per explant. The most suitable sugar concentration was30g/1. The optional time for the culture was35days.4. Hongyan1plants were able to grow normally in the test area. In the test, Hongyan1grew normally andits fruit finger length was long and the comb was nice. Hongyan1had a higher productionand a better fruit quality.5. In the process of banana planting, the controlled release fertilizer could save much labor so as to achieve a good effect as well as a better pseudo-stem height. |
