Musa nana Lour, belongs to Musa, Musaceae. Most kinds of the Musa nana Lour are triploid. So it is difficult to breed them by traditional methods, for example, crossbreeding.The traditional way for breeding Musa nana Lour is bud-reproduction. But if things go continue in this way, the seed nature of the Musa nana Lour may become worse and worse. So, it is in poor state for us to choose the new breeds of Musa nana Lour with fine properties. Using biotechnology may break these limitations. The research includes the experiment about how to lead different explants become embryogenic callus, how to establish the embryogenic cell suspensions, and how to regenarate the embryogenic callus into banana shoots. In these experiments, we systematically study the process, techniques and influencing factors of the regenaration. Main achievements of our study are as below:①The induction of the Musa nana Lour’s cauloid form into the embryogenic. When we induced the cauloid of dwarf banana, the culture medium of T2(MS+TDZ0.02mg/1+2,4-D7mg/1) had the highest inductivity, as the value was9%; when induced the Hongyan1, the culture medium of T4(MS+TDZ1.0mg/1+2,4-D7mg/1) had the best inductivity, as the value was3%; when induced the Guijiao6, the culture medium of T2had the highest inductivity, as the value was6%.②The induction of the different parts of the dwarf banana into embryogenic callus:the most appropriate part for the induction was petiole, the culture medium which wad the most appropriate was T21(MS+TDZ0.2mg/l+2,4-D5mg/l). The inductivity was the highest, up to63%. ③The induction of the banana flower bud to regenerate embryogenic callus:As study to the Guijiao6banana the best part for inducing was the top part of the flower bud, M1(MS+VH lmg/1+NAA lmg/1+IAA lmg/1) was the most appropriate culture medium, as the induction rate reached43,8%. When researched to the flower bud of the dwarf banana, the most appropriate part for inducing was middle part. And the best culture medium was M1, as the inductivity was highly to66.7%.④The establishment of the ECS:It was much more easy for dwarf banana to establish the ECS with the embryogenic callus form from the cauloid. The most appropriate liquid medium was ZT2(VC10mg/1+2,4-D1.lmg/1+ZT25mg/1).⑤The experiment of the shoots regeneration:The flower bud of the Guijiao6banana had the highest inductivity, as the rate was81.8%. The best medium for regeneration was Y3(MS+ZT0.05mg/1+TDZ0.16mg/1+NAA0.2mg/1).
