This study aims to culture conditions through the optimization of fermentation of probiotics, and make it into a high activity compound bacteria preparation, used in pig production. Determination of growth curve of each test strain of the test, the unit number of live bacteria as index, the optimal design and screening of probiotic fermentation time and the optimal medium composition. Then the laboratory screening of6strains of probiotics to obtain solid compound bacteria preparation were used in lactating sows, piglets, and, provide the theoretical basis and application of parameters for the application of probiotics in pig production..Test1Through using the methods of plate colony counting, determination of growth curve of Lactobacillus acidophilus, and found that the stable growth period appeared in20-22h; the fermentation time and glucose, sucrose, peptone, yeast extract, citric acid salt of5kinds of optimization of medium components, determine the optimal fermentation time of acidophilic lactobacillus as20h, for the optimal fermentation medium components:glucose110g/L,10g/L sucrose, peptone5g/L, yeast extract30g/L,12g/L citric acid salt.Test2through the determination of growth curve of Bacillus coagulans and thermophilic Bacillus stearothermophilus mixed culture, that the growth of Bacillus appears at30h; the fermentation unit living bacteria number index to measure the judgment formula three is the best medium, the optimal medium composition:80soybean powder1%,80wheat bran powder1%,80orders,1%corn powder, glucose1%, CaCO30.3%, K2HPO40.2%, MnSO4.H2O0.002%, MgSO4.7H2O0.01%, NaCl0.1%, H2O100mL. Bacteria living bacteria number up to1.95Ă—1012cfu/mL.Test3selected the30d before delivery, birth weight, birth similar purebred Landrace sows, feeding test requirements were randomly divided into experimental group and control group2treatments,2replicates per treatment, each repeated7sows, the test group fed with basal diet plus0.2%composite probiotics, blank control group fed with basal diet, feeding time from prenatal since30d delivery. The results showed that, the survival rate of piglets, the test group than the control group is3.49%, difference was significant (P<0.05); the two group of average litter weight at birth and the average birth weight, the difference was not significant (.P>0.05). The experiments show that prenatal sows fed the composite probiotics, can significantly improve the survival rate of piglets. Test4composite probiotics into suckling and weanling piglets diets according to the proportion of0.2%, and the control of feeding trials with antibiotics group. Body weight, litter size, birth order, birth date is similar to sow purebred Landrace piglets by feeding60head, were randomly assigned into2treatments,3replicates per treatment,10piglets per replication, were fed antibiotics (antimycin, aureomycin, with Jane Kui Yichun) and two kinds of dietary compound probiotics preparation. The average of two groups of piglets lactation, weaning feed intake, average daily gain weight per head difference, total feed intake, feed conversion ratio, diarrhea rate and mortality were not significant (P>0.05).The experiment results:the maternal effect of probiotics can improve the survival rate of piglets after weaning, improve the disease resistance, control of intestinal diseases, promote the growth of piglets. Composite probiotics can substitute the antibiotics in a certain range.
