In order to private sufficient high-quality pronuclear embryo for transgenic microinjectionand improve the efficiency of pronuclear embryo production,128goats were selected as donors(1-5years old). The embryos were transferred to192receptor goat after DNA microinjection.The effect of breeds (Jia Xiang goat and Yi Shui goat) in Shan Dong, season, weight, number ofoperation on superovulation combined with endoscope semen were studied by analyzing theresult got from experiment. The influence of breed, season, weight, number of operation and thenumber of embryo transferred on pregnant rate was concerned as the same time. Correspondingoptimization suggestions were put forward. Result shows that the production efficiency ofpronuclear embryo of two breeds is similar; when the surgery number less than or equal to3,superovulation efficiency was not influenced significantly by surgery times (p>0.05). When itcomes over3times, the number of Embryos decrease with the surgery times increased tosignificant difference (p<0.05). FSH8injection slowing reduction method in combination withendoscopic semen method can get more embryos in pronuclear period (90.21%) compared withnatural breeding. Significant higher embryo production efficiency got from donor weightedover40kg rather than30-40kg (p<0.05), while higher pregnant rate got from the receptorsweighted over50kg. The production efficiency of donors and receptors was not influenced byseason(P>0.05). The goat pregnant rate raised with number of embryo they receipted increased,but not come to significant (P>0.05). Above all, choosing goats which are under3times surgeryand over40kg as donors with FSH8injection slowing reduction method with endoscopicsemen method can improve the efficiency of pronuclear embryo production.3pronuclearembryo transfer to receptors which weighted over50kg can optimize utilization rate ofembryos. The goat pregnant rate rose with number of embryo which goat receipted increased.
