| The research on the cultire of Pleurotus eryngii, a kind of excellent quality rare fleshyagarics, and in order to promote the healthy and steady development of the P. eryngiiindustry.The influence of temperature on P. eryngii growth process and extracellular enzymeactivitis, trehalose production and trehalose synthase activity in Pleurotus eryngii at differentdevelopment stages are studied in this paper.1. The strain Pleurotus eryngii ACCC52627provided by Agricultural Culture Collection ofChina, cultured at17°C,19°C,21°C,23°C,25°C,27°C,29°C,31°C, and recorded the myceliumgrowth rate and growth vigor. After full growth of mycelia, cultivated experiments wasperformed, and growth cycle, yield andbiology efficiency were recorded, through acomprehensive comparative analysis concludes as follows: mycelium growing well under theconditions of23°C and25°C, the substrates are coverd with mycelium used less days andmycelial average growth rate of about3.5mm/d or so, a short cultivated period, fruiting yieldhigher and the average yield is up to310g or more, biotransformation rate as high as about90%,the time of harvesting is relatively concentrated, better uniformity; and the mycelium cultured in23°C had shorter cultivated periods, and the average time of fruiting is about15d; the optimumtemperature for mycelium culture stage is23°C.2. The results of the influence of temperature on extracellular enzyme activitis in P. eryngiiat mycelium cultured periods showed that: extracellular enzyme activitis changes throughout themycelium culture process has obvious periodic steps, and the temperature had somehow effectson the extracellular enzyme activitis. Activity changes of cellulose and hemiceluase are verysimilar, the activitisare low oninitial training, with the continuous growth of the mycelium,enzyme activitis level gradually increased, reaching a maximum value at after-ripening; theamylase activity level wasinstability throughout the whole phase of mycelium culture, and thereis no significant influence of temperature on it.3. The content of trehalose in P. eryngii was determined by high performance liquidchromatography (HPLC), and the influence of temperature on trehalose content was studied.The results showed that: trehalose content changes law throughout the growing period appearsobvious phases, and reached a peak at the formation of primordium; seen from the culturetemperature, trehalose content in mycelia gradually increases with the rise of the temperature within the temperature range for normal growth of Pleurotus eryngii mycelia, has no obviousincrease at19-25°C, and has sharp increase when the culture temperature is31°C, whichexceeding normal culture temperature and mycelium growth is under stress. After thetemperature reached in37°C, mycelial growth is stopped, the content of trehalose is rapiddecline in the mycelium.4. DNS method and differential methodwere adopted, in the determination, reducing sugarin the system was determined to indirectly calculate the content of trehalose from enzymereaction, thereby reflecting the activity of trehalose synthase, results showed that: The change ofthe extracellular trehalose synthase activity in the whole growing period is similar with that ofthe trehalose content. But trehalose synthase activity is not the highest onformation ofprimordium and fruiting buds phases, although the trehalose content at a higher level. |
