Rice is one of the main food crops in China, more than50%of the Chinese made rice asthe staple foods. Developing the two-line hybrid rice is imperative to expand the cultivated areaof hybrid rice and increase grain production. It has important significance to study the breedingof PTGMS line in rice and the research of mechanism of fertility restoring and the geneticprinciples. That is because the PTGMS line in rice is the foundation to develop the two-linehybrid rice. This research records the breeding procedure of PTGMS line in rice, the alterationrule of PTGMS line in rice and the molecular detection. The results show that:1. According to the breeding program of PTGMS line in rice, bred through the cross of DS withstability and L39with photo and thermo insensitive conventional rice varieties. We can getPTGMS Line L39S with good characters and stable fertility through breeding wish shuttle backand forth from Hainan to Tianjin.2. The combinations with the same background isolated different ratios in Liaoning, T Tianjincity and Jiangsu Province. According to the survey of the day length and temperature in thethree locations. Most of the sterilities are thermo insensitive isolated from F2population underthe environmental conditions in Liaoning, the photo insensitive isolated from F2populationunder the environmental conditions in Jiangsu. The thermo insensitive, photo insensitive andphoto-thermal interaction sterilities are all isolated under the environmental conditions inTianjin. That is because the full expression of the sterile character under the environmentalconditions in Tianjin.3. Although the photo-thermo-sensitive genic male-sterile gene of rice has been located by SSRmolecular marker, but the PTGMS lines with different genetic background have different sitesto control the fertility in rice. Using the SSR markers has been the located cannot locate thephoto-thermo sensitive genic male-sterile gene in L39S, L62S, DS. So develop the new primersand locate position is necessary if you want to use SSR molecular marker to locate the threesterile lines. Using the SSR markers can not explain the phylogenetic relationships betweenPTGMS lines in rice.4. Using the SNP molecular marker technology to detective the photo thermo sensitive genicmale sterile rice and normal fertile rice varieties. The photo thermo sensitive Genic Male SterileRice SNP locus carrying a thermo sensitive genic male sterile gene p/tms121for G, threenormal fertility of rice variety p/tms121the SNP site is C, this result and the Zhou result is consistent. Three place DS/Jiamusi-2combination of F2generation male sterile plants andfertile plants were SNP, found that p/tms12-1sites affected by environmental regulationeffects of severe environment in Tianjin, the critical value and fluctuation, so the expression ismost fully. However, there are still mutant in sterile plants, control so the author inferencetwo-line sterile line but also by other major gene or polygenic.
