Chinese fir(Cunninghamia lanceolata) is the most important tree species in South China. This plantation has an important position in China.Open tissue culture plants referred to that under the effect of antibacterial agent,plant tissue culture does not require strict aseptic operation environment and high pressure sterilization as well as clean bench,It can carry on the plant tissue culture under the opening of bacteria in the environment,So the open tissue culture radically simplify links,reduce the cost of tissue culture,meet a real industrial requirements.This research will apply the open tissue culture on the pine, aimed at exploring and establishing a " Open tissue culture " mode,radically simplified Chinese Fir tissue culture links,reduce the cost of tissue culture.so that the tissue culture can really be used for the production of Chinese FIR and serviced in agriculture,for the realization of culture opened up a new road of industrialization.In the process Open tissue culture for Chinese Fir,we have filtered the bacterio static agent and have measured the plant height,fresh weight,dry weight,the number of root,the root length and chlorophyll fluorescence parameter of the Chinese Fir which grow on the agent added antibacterial.It comes to the conclusion as follow:(1) Through the separation and purification of air pollution in fungi,received 9 species of fungi,such as:2 species of Aspergillus,2 species of Mucor 2,3 species of Penicillium,Trichoderma and Rhizopus.Comparison of the effect of antibacterial different concentrations of antimicrobial agents derived,the effect of antibacterial Trimangol is most obvious.80% mancozeb have some inhibitory effect on 5 species of fungi.The highly concentrated sodium hypochlorite has the very strong inhibitory action to these 5 kind of fungi,but the low concentration (0.1%) piece of function is small. The antibacterial effect of carbendazim and chlorothalonil is not ideal, Rhizopus and Mucor of no effect.(2) Exam the inhibitory action of several kind of mix bacteriostats to the group the common fungus category in the process of tissue culture.finally obtains the No.9 which 62.5mg/L80% mancozeb and 62.5mg/L Trimangol and 0.5% sodium hypochlorite this mix bacteriostat the bacteriostasis effect to be best,may suppress the all laboratory cmmon fungus category nearly.Guang Puxing is good.After joins the combination 9 mixed Antimicrobial agent , carries on the culture medium configuration under the opening condition which the pollution rate is only 3.4%,as same as the culture medium which carried on under strict high-pressured,but the pollution rate of the culture medium which has not joined the bacteriostat achieved 27%. therefore the effect of this kind of mix bacteriostat is very obvious. (3) The effect of antibacterial agent for seedling growth,after with the combination 9 the growth of fir is in good condition.Through the variance results and multiple comparisons,At the the height,fresh weight and dry weight of the fir,A large concentration of combination 1 against the control group is not remarkable ,while the concentrations 9 against the control group is remarkable;It can be inferred from the chlorophyll fluorescence kinetics parameter that the combination 9 of a small concentration has little impact on the synthesis of chlorophyll fir and photosynthetic capacity,even compared to the impacts exerted by the control group;Regarding the growth condition of fir root in open tissue culture,through the variance results and multiple comparisons,it can be noticed that the inhibitory concentration has little effects on the fir root amounts but weigh much in the length of the root.In similar to the results found between the inhibitory concentration and the growth condition of seedlings,the inhibitory concentration is also in direct proportion to the length of root.that is to say,the inhibitory concentration is in direct proportion to the growth condition of seedlings.Hencefore,the seedling which is added with the combination 9 of the smallest concentration,has the best condition and has the slightest difference with the control group.(4)The open tissue culture system of Chinese Fir have been established,in the open tissue culture of Chinese Fir,Except the bacteriostatic agent,the environment and the technology of requirements are aslo important,only according to the requirements of tissue culture system strictly,the open tissue culture system of Chinese Fir will be established successful.(5)The analysis of comparison on the cost of open tissue culture and tradition tissue culture , obtains the cost of open tissue culture to be lower. Fir seedlings showed the cost of per plant decreased has decreased by about 30%, which would save a lot of fir seedlings factory production costs.Moreover, the absence of equipment limitations,the establishment of tissue culture plants will be more simple. At the same time open tissue culture without high pressure sterilization,thus saveing a lot of time and Manpower,so open tissue tissue culture have huge advantages and prospects for the shortage of time and human resources in modern economic.
