In recent years, salt-stress caused by the salinization of soil is a main environmental factor restraining plant growth and yield, which severely restricts the development of modern agriculture. The aim of this paper is to find new wheat salt-tolerant genes transformed into Arabidopsis thaliana and analysis salt tolerance of these transgenic plants. The main results of this article were:1. functional analysis of the gene named TaPRKc in WheatOn the basis of under-graduated sister who found a gene named AtKPP usinng the At2g32500 in Arabidopsis thaliana that has a high homologous with TaSR in Wheat by two-hybrid system in the cDNA library of Arabidopsis thaliana. Have been known the interaction KPP and PRK in the tomato. The NO. of PRK been known by the reports. Obtained the sequences of TaPRKc in Wheat by blasting on the ncbi web page. Bioinformatics of the gene has a PKc-like superfamly conserved domain.To study the function and its role of TaPRKc gene in salt stress, TaPRKc was subcloned into eukaryotic expression vector and transformed into Arabidopsis thaliana. Treating with NaCl of different concentrations, survival rate of homozygous transgenic lines were less than that of the wildtype plants. It suggests that TaSR gene could not enhance salt tolerance.2. Functional analysis of drought-resistant TaSR in wheatThe relative expression of TaSR was detected by real-time PCR under PEG stress. The expressions of this gene were higher in wheat halotolerant mutant than that in salt-sensitive mutant under PEG treatment. The expression of the gene was significantly high in the roots and leaves under PEG stress. So it may be related with PEG tolerant.TaSR was subcloned into eukaryotic expression vector and transformed into Arabidopsis thaliana. Treating 8%PEG, root length, transgenic plants are better than the wildtype plants and mutant plants. it suggests that TaSR gene could improve drought tolerance of transgenic Arabidopsis thaliana.
