Application Of The Colloidal Gold Immunochromatographic Test Strips For Residues Detection Of Sulfonamidesin Aquatic Products

In this study,the rapid detection of sulfonamides residues was established by Colloidal gold immunochromatographic test strips.The major results are as follow:1. Triplet-colloidal gold strip of sulfamethazine, sulfadiazine-sulfaquinoxaline and penta-colloidal gold strip of sulfamethazine, sulfaquinoxaline, Sulfamonomethoxine, Sulfametoxydiazine and Sulfadimethoxinewere prepared using the method established in our laboratory.All the sensitivity of detecting these six sulfonamideswas 100ng/ mL, consistent, withnational dose limited standard, and it could be used in detection of multi-sulfonamides residue.2.The pretreated method for sulfonamides in fish was developed. Based on the standard curves, rate of recovery was from 70% to 95% for six sulfonamidesin fish, and coefficient of variation (CV) in different lot was less than15%.3.Fish feeding with the therapeutic doseof three different sulfonamides(SD, SM2 and SMM)everyday for 5days respectively, sulfonamides residues in fish with feedingdrugs werepositive detected by test stripsin the first day of feeding,but it was negative in fish without feeding drugs. The residues of SD and SM2in fish still could be detectedin the first 5-6 days after stopping feeding drugs; the residue of SMMcould be converted to negative after 3 days, because the metabolism of SMMwasrapid.A comparison between the test strips and ELISA was performed with fish samples from the animal experiment. The positive coincidence rate was 86.6%, the negative coincidence rate was 86.6%, and the total coincidencerate was 92.9%.The results indicated that all theimmunochromatographic rapid test strips can meet the needs of screening sulfonamides residues in fish. In conclusion,it could be used in the detectionof sulfonamids residues in aquaticproducts.
