| Sulfonamides are widely used in animal husbandry and as feed additives. As a result, food derived from animals treated with sulfonamides may be contaminated with those drugs. The medcines will enter human body and eclogical system by food chain, and will do harm to our health at the same time pollute the invironment. Its influence our animal production exporting seriously, and make a big lost in economy. So it is very important to strengthen the using of ainimal medcine and testing works.Nowadays, HPLC is the authorized standard method for testing sulfonamides. There are many reports about the residuing of sulfonamides by HPLC internal and overseas. But because there are so many kinds of sulfonamides, the method today can't aim directly at all of them. So my experiment object is to establish a method which can test many kinds of sulfonamides residuing in ainimal food at the same time to be living on the research that the predecessor made. So this method will supply for testing sulfonamides residuing in ainimal food quickly by Veterinary Health Superintend Department,Farm Products Mass Detection Department,Livestock Genicing And Processing Department,The Food Hygiene Testing Department and so on. And then we can direct the rational administration, test and control the residuing of sulfonamides in ainimal food, and so we can protect our health.In the study about establishing the HPLC condition, SD, SA, SQ, SUL, SMM, SMD, SDM', SIZ, ST, PST, SST and SMZ by ultraviolet were scaned respectively. The result shows that sulfonamides obsorb better at about 270nm. So we choose the wave length at 270nm. Because of the different acidity or alkalinity and formation among sulfonamides, we use different mobile phase.We choose acetonitrile and aceticaid for mobile phase, then elute sulfonamides gradiently in order to separate sulfonamides in short as possible as we can.To fumble about the condition of sample treatment, we use acetonitrile- chlorlform as extract. There are little impurity in this method, and extract SDM'and SMZ, PST, SSTand ST can't separate. Recovery rate are above eighty percent.In the part of feeding and injecting animals with sulfonamides, we can't test SMD and SMZ after the feeding and sample processing. We doubt that sulfonamides were acetylated or oxidized to other kinds of sulfonamides. They can't appear on former location. SQ appear on former location both in it's nomal dos and dosis toxica. The peak form and peak hight both well. So we can conclude that SQ residues acutely. SUL is as same as SQ. Because of that, we should abide by direction and dosage when we use sulfonamides, otherwise it will do harm to our health if we eat the animal food with residue of sulfonamides.Beside, we also do the blood biochemistry analyze experiment. The result shows that SQ influences AST, ALT, TP, CHO and LDH severely in it's nomal dosage (P<0.05), and influences ALP, ALB and UA unacutely in it's nomal dosage. SMZ influences ALP and UA acutely in it's nomal dosage (P<0.05). And it influences ALP, TP and CHO markly in it's toxic dosage (P<0.05). |
